Description

Apaf-1, also known as apoptotic protease-activating factor 1, is a protein that plays a critical role in inducing apoptosis or programmed cell death. It mediates the cytochrome c-dependent activation of pro-caspase-9, which ultimately leads to the activation of caspase-3 and the execution of apoptosis. It is important to note that this process requires ATP.
There are six isoforms of Apaf-1 that are produced through alternative splicing of the gene. However, isoform 6 is less effective in inducing apoptosis than the other isoforms.
The activation of Apaf-1 is induced by E2F and p53 in apoptotic neurons. When cytochrome c binds to Apaf-1 and ATP, it leads to the oligomerization of Apaf-1. Pro-caspase-9 then binds to the Apaf-1 oligomer via their respective NH2-terminal CARD domains. This binding leads to the autocatalytic activation of pro-caspase-9 and subsequent release of mature caspase-9 from the complex.
Pro-caspase-3 is recruited into the Apaf-1-pro-caspase-9 complex via interaction with pro-caspase-9. The activation of caspase-3, which is a key player in the execution of apoptosis, ultimately leads to cell death.
It is also worth mentioning that Apaf-1 interacts with APIP, an apoptotic protein, which further regulates the apoptotic process. Overall, the crucial role of Apaf-1 in regulating apoptosis makes it an important target for therapeutic interventions in various diseases such as cancer.
