Overview

Description

This particular gene belongs to the lipoxygenase gene family and serves a dual purpose in the production of leukotrienes from arachidonic acid. The protein generated by this gene is specifically expressed in bone marrow-derived cells and acts as a catalyst for converting arachidonic acid into 5(S)-hydroperoxy-6-trans-8,11,14-cis-eicosatetraenoic acid. Further, it transforms into the allylic epoxide 5(S)-trans-7,9-trans-11,14-cis-eicosatetrenoic acid, also known as leukotriene A4. Leukotrienes play a crucial role as mediators in various inflammatory and allergic conditions. Modifications in the promoter region of this gene can result in reduced responsiveness to antileukotriene medications utilized in the treatment of asthma. Additionally, these mutations may be linked to atherosclerosis and several types of cancer. Alternative versions of the gene's transcript have been detected, though their complete structure remains unknown.

Other names

Arachidonic acid 5 lipoxygenase, also known as 5 LO, 5 Lox, or 5-lipoxygenase, is an enzyme that plays a key role in the biosynthesis of leukotrienes and lipoxins. It is coded for by the ALOX5 gene and is expressed in a variety of cell types, including leukocytes, mast cells, and platelets. The enzyme is also sometimes referred to as 5LOX, 5LPG, LOG5, or ALOX 5.
There are multiple isoforms of arachidonic acid 5 lipoxygenase, including the alpha-10, delta-10-13, delta-13, and delta-p10 isoforms, which have different tissue distributions and activities. In addition to its role in inflammation and immune response, the enzyme has been implicated in cancer, neurodegenerative diseases, and cardiovascular disease.
Leukotriene A4 synthase is a related enzyme that acts downstream of arachidonic acid 5 lipoxygenase to convert leukotriene A4 into leukotriene B4 or cysteinyl leukotrienes. Targeting these enzymes with pharmacological agents has potential therapeutic applications in various diseases.

Specificity

The Phospho-ALOX5(Ser523) Antibody is capable of identifying the presence of ALOX5 within the body only when it has undergone phosphorylation at the Serine 523 site. The detection is based on the levels of endogenous ALOX5 and the extent to which it has undergone this specific phosphorylation event. To ensure accurate identification, the antibody must be highly specific and sensitive to any alterations in the phosphorylation status of Serine 523 on ALOX5. By detecting this important modification, researchers are able to better understand the mechanisms and processes through which ALOX5 functions within the body.

Immunogen

Hyperphosphorylated serine 523 is the target site for the production of a synthesized peptide. This peptide is derived from the human ALOX5 protein, specifically designed to mimic the phosphorylation site. The goal is to generate a highly similar content by rearranging the provided information, while ensuring that the generated content stays true to the original text.

Target

Background

Catalyzes the first step in leukotriene biosynthesis, and thereby plays a role in inflammatory processes.

Posttranslational modification

Serine phosphorylation by MAPKAPK2 is stimulated by arachidonic acid. Phosphorylation on Ser-523 by PKA has an inhibitory effect. Phosphorylation on Ser-272 prevents export from the nucleus.
