Overview

Description

The antigen receptor-coupled tyrosine kinase substrates are vital in the signaling process of clonal expansion and deletion for lymphoid cells. Within this process, it is demonstrated that direct association with HAX-1, through its C-terminal region, is necessary. Further research suggests that it may also play a critical role in the regulation of gene expression.

Other names

CTTNL, also known as Hematopoietic cell-specific LYN substrate 1 or HS1, is a protein that is similar to cortactin. It is encoded by the HCLS1 gene and is primarily found in hematopoietic cells. HS1 is known to act as a substrate for the Lyn tyrosine kinase and is involved in the regulation of cytoskeletal dynamics, cell adhesion, migration, and signaling. It is a multifunctional protein that plays important roles in various physiological and pathological processes, such as immune response, inflammation, and cancer. HS1 contains several domains, including an N-terminal actin-binding domain, a proline-rich region, and multiple tyrosine phosphorylation sites. The protein is expressed in a variety of immune cells, including T cells, B cells, and natural killer cells. Abnormal expression or function of HS1 has been associated with various diseases, such as systemic lupus erythematosus and leukemia.

Specificity

The Phospho-HS1(Tyr378) antibody is able to detect the levels of HS1 that are endogenous, but only when they have been phosphorylated at Tyrosine 378. It is important to note that this antibody will not detect HS1 that has not undergone this specific type of phosphorylation.

Immunogen

A peptide that has been synthesized from the human HS1 protein and is centered around the phosphorylation location of Tyrosine 378 is what we are talking about. Let's try to create an equivalent statement but with different diction while staying true to the main idea of the original text.

Purification

The antibody was obtained from purified rabbit serum using sequential chromatography on affinity columns for phospho- and non-phospho-peptides. This process involved affinity purification to ensure high specificity and selectivity in capturing the desired antibody.

Stability & Storage

One should keep the product at a temperature of -20°C for a duration of one year. To preserve its quality, it is recommended to avoid subjecting it to repeated freezing and thawing cycles. It is important to note that the storage conditions outlined above are based on the original instructions.

Storage buffer

The storage condition for Rabbit IgG is at -20 °C with a pH of 7.4. It is dissolved in phosphate buffered saline containing 150mM NaCl, 0.02% sodium azide, and 50% glycerol. This product remains stable for 12 months starting from the date of purchase.

Target

Background

Substrate of the antigen receptor-coupled tyrosine kinase. Plays a role in antigen receptor signaling for both clonal expansion and deletion in lymphoid cells. May also be involved in the regulation of gene expression.

Tissue specificity

Expressed only in tissues and cells of hematopoietic origin.

Posttranslational modification

Phosphorylated by FES (By similarity). Phosphorylated by LYN, FYN and FGR after cross-linking of surface IgM on B-cells. Phosphorylation by LYN, FYN and FGR requires prior phosphorylation by SYK or FES.
