Overview

Description

CCR5 is a receptor that belongs to the 7-transmembrane G-linked receptor family. It specifically binds to various inflammatory C-C chemokines such as MIP-1-alpha, MIP-1-beta, and RANTES. Upon activation, it transduces a signal by increasing the levels of intracellular calcium ions. This receptor may have a role in regulating the proliferation and differentiation of granulocytic lineage cells. Additionally, it acts as a co-receptor, along with CD4, for HIV-1 R5 strains and interacts with the viral surface protein gp120. The efficient binding of ligands CCL3/MIP-1alpha and CCR4/MIP-1beta requires specific modifications like sulfation, O-glycosylation, and sialic acid. Notably, glycosylation on a specific site, S6, is necessary for the effective binding of CCL4.

Other names

CCR5, also known as CC chemokine receptor type 5 or C-C chemokine receptor type 5, is an antigen present in humans. It plays a crucial role as the HIV-1 fusion coreceptor and is associated with various diseases such as IDDM22. Other names for CCR5 include CKR5, CD195, CMKBR5, and CHEMR13. It is also referred to as CCR5 chemokine receptor or CC chemokine receptor type 5. Additionally, it is sometimes denoted as CCR5_HUMAN or FLJ78003. This receptor is responsible for binding with chemokines like MIP-1 alpha and functions as a receptor for them.

Specificity

The presence of phosphorylated CCR5 at Serine 349 can be identified using the Phospho-CCR5(Ser349) Antibody. This antibody specifically detects endogenous levels of CCR5, but only when it is phosphorylated at Serine 349. By targeting this phosphorylation site, the antibody allows researchers to study and understand the role of phosphorylated CCR5 in various biological processes.

Immunogen

A peptide that has been synthesized from human CCR5 and is focused on the serine 349 phosphorylation site, can be derived. To create similar content, the main idea is to emphasize the peptide, human CCR5 and Sersine 349 phosphorylation site. Thus, one can mention that the peptide has been synthesized from human CCR5, with emphasis on Sersine 349 phosphorylation site.

Target

Background

Receptor for a number of inflammatory CC-chemokines including MIP-1-alpha, MIP-1-beta and RANTES and subsequently transduces a signal by increasing the intracellular calcium ion level. May play a role in the control of granulocytic lineage proliferation or differentiation. Acts as a coreceptor (CD4 being the primary receptor) for HIV-1 R5 isolates.

Tissue specificity

Highly expressed in spleen, thymus, in the myeloid cell line THP-1, in the promyeloblastic cell line KG-1a and on CD4+ and CD8+ T-cells. Medium levels in peripheral blood leukocytes and in small intestine. Low levels in ovary and lung.

Posttranslational modification

Sulfated on at least 2 of the N-terminal tyrosines. Sulfation contributes to the efficiency of HIV-1 entry and is required for efficient binding of the chemokines, CCL3 and CCL4.O-glycosylated, but not N-glycosylated. Ser-6 appears to be the major site. Also sialylated glycans present which contribute to chemokine binding. Thr-16 and Ser-17 may also be glycosylated and, if so, with small moieties such as a T-antigen.Palmitoylation in the C-terminal is important for cell surface expression, and to a lesser extent, for HIV entry.Phosphorylation on serine residues in the C-terminal is stimulated by binding CC chemokines especially by APO-RANTES.
