Overview

Description

Mouse Monoclonal Antibody (Mab)

Other names

Pin1, also known as peptidyl-prolyl cis-trans isomerase NIMA-interacting 1, is a protein that plays an important role in regulating cellular processes. This enzyme, which belongs to the family of peptidyl-prolyl cis-trans isomerases, is responsible for catalyzing the isomerization of specific proline residues in target proteins.
Pin1 is involved in a wide range of cellular functions, including cell cycle progression, DNA damage response, and transcriptional regulation. It has been shown to interact with a variety of protein targets, including kinases, transcription factors, and cell cycle regulators, and to modulate their activity through post-translational modifications.
One of the most important functions of Pin1 is its ability to regulate the activity of oncogenic proteins. Pin1 has been shown to promote the degradation of tumor suppressor proteins, as well as to enhance the activity of oncogenic proteins. This suggests that Pin1 may play a critical role in the development and progression of cancer.
Overall, Pin1 is a highly important protein that is involved in a number of key cellular processes. Further research into the mechanisms of Pin1 regulation and activity may lead to new therapeutic targets for the treatment of cancer and other diseases.

Mouse

Specificity

This monoclonal antibody was generated using purified His-tagged PIN1 protein. The antibody was specifically developed to identify and bind to the target protein with high affinity and specificity. The production process involved careful selection of hybridoma cells that secrete antibodies against the PIN1 protein, followed by purification and characterization of the final product. The antibody is expected to have a wide range of applications in various research fields, particularly in studies involving PIN1 protein function and regulation.

Stability & Storage

To ensure proper storage, it is recommended to keep the refrigerated temperature between 2-8°C for a maximum of 2 weeks. If you need to store it for a longer period, it is advisable to freeze it at -20°C in smaller portions to prevent repeated freeze-thaw cycles.

Storage buffer

The provided monoclonal antibody is supplied in PBS containing 0.09% (W/V) sodium azide. To obtain this purified antibody, a protein G column was used for purification. It was then eluted using high and low pH buffers and promptly neutralized. Subsequently, the antibody underwent dialysis against PBS to ensure its purity.

Background

This protein, which is essential for mitosis regulation, is believed to interact with NIMA and reduce its activity in promoting mitosis. It specifically prefers an acidic residue located before the isomerized proline bond. Its main function is catalyzing the cis/trans isomerization of pSer/Thr-Pro bonds. Additionally, it has the ability to decrease the kinase activity of BTK. Furthermore, it has the ability to activate multiple oncogenes, leading to centrosome amplification, chromosome instability, and cell transformation. Moreover, it plays a crucial role in efficiently dephosphorylating and recycling RAF1 following mitogen activation. It also binds to PML and BCL6, targeting them for degradation in a phosphorylation-dependent manner.

Celluar localization

Nucleus. Nucleus speckle. Cytoplasm. Note=Colocalizes with NEK6 in the nucleus. Mainly localized in the nucleus but phosphorylation at Ser-71 by DAPK1 results in inhibition of its nuclear localization
