Overview

Description

The KRT7 gene encodes the CK7 protein, a type II cytoskeletal keratin found in humans. It is predominantly expressed in the simple epithelia that line the internal organs' cavities, as well as in gland ducts and blood vessels. CK7 is part of the keratin family, which includes neutral and basic proteins forming heterotypic chains during the differentiation of simple and stratified epithelial tissues. This specific type II cytokeratin is localized in the aforementioned tissues, and its encoding genes are situated in a region on chromosome 12q12-q13. Several transcript variants may arise from alternative splicing, although not all have been fully characterized.

Other names

Cytokeratin 7, also known as CK-7, is a type of keratin protein that is commonly found in simple epithelial cells. It plays an important role in maintaining the structural integrity of cells, and is involved in a wide range of cellular processes. CK-7 is encoded by the KRT7 gene, and is also referred to as keratin type II cytoskeletal 7. It is often used as a marker for the differentiation status of cells, and can be detected using immunohistochemistry techniques. In addition to its role in cellular function, CK-7 has been implicated in various diseases, including cancer. Researchers are currently exploring its potential as a diagnostic and therapeutic target in these conditions.

Specificity

The detection of total CK7 is possible using the CK7 antibody, which specifically targets endogenous levels of this protein. Rearranging the information from the original text, the CK7 antibody has the ability to recognize and bind to all forms of CK7 present within the sample being analyzed. This allows for accurate identification and quantification of total CK7 levels in various biological systems.

Application

ELISA is a commonly used technique in immunology to detect the presence of specific antibodies or antigens in a sample. The dilution used for ELISA is typically 1 in 10,000, indicating that the sample is diluted 10,000 times before it is added to the assay.
Western blotting (WB) is another widely used technique to detect specific proteins in a sample. The dilution range for WB is usually between 1 in 500 and 1 in 2,000, meaning that the sample is diluted accordingly before being loaded onto the gel for electrophoresis.
Flow cytometry (FCM) is a powerful method for analyzing and characterizing cells in suspension. The recommended dilution for FCM analysis is generally 1 in 200 to 1 in 400, indicating that the sample is diluted appropriately with buffer solution before analysis.
ELISA using peptide antigens is a variation of the standard ELISA technique. The recommended dilution range for ELISA with peptide antigens is usually between 1:20,000 and 1:40,000, indicating that the sample is diluted accordingly before being added to the assay wells.
It is important to note that these dilution factors are guidelines and may vary depending on the specific experiment or assay conditions. It is recommended to optimize the dilution for each experiment to ensure accurate and reliable results.

Immunogen

A fragment of human CK7 was expressed in E. Coli and then purified for use. The recombinant fragment closely resembles the original version, as it was produced using advanced techniques to ensure its accuracy and completeness. This purified material is of great value in research and medical applications, given its high level of similarity to naturally occurring CK7. Thanks to its purity and quality, this recombinant fragment has become an important tool for studying CK7 and related proteins, as well as for developing new treatments and therapies. Overall, this purified fragment represents a major advance in our ability to understand and manipulate this important protein.
