The Interleukin-17 family encompasses six proteins (IL-17, IL-17B through IL-17F) that share a similar structure and the ability to induce local cytokine production. While the C-terminus contains a conserved cysteine-knot fold, there is considerable divergence at the N-terminus. IL-17D is the most closely related to IL-17B with a 27% sequence homology. It is predominantly expressed in tissues such as skeletal muscle, heart, adipose tissue, lung, pancreas, and nervous system. As with other members of the IL-17 family, IL-17D modulates immune responses indirectly by stimulating the production of myeloid growth factors and chemokines including IL-6, IL-8, and GM-CSF.
