Testing Principle：

This ELISA kit utilizes the double antibody sandwich technique, with a prepackaged high affinity conjugate plate containing the specific anti-human CXCL13 Capture Antibody. The sample to be tested, standard substance, and biotin labeled detection antibody are sequentially added to the HRP-plate well for complete oscillation and mixing. The sample is then incubated at room temperature for two hours to bind CXCL13 to the solid phase antibody and detection antibody. After thorough washing to remove unbound ingredients, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is added followed by another wash cycle. TMB color substrate is then added and incubated in the dark at room temperature for color development, with the depth of color reaction positively correlated to the CXCL13 concentration in the sample. The reaction is stopped by adding the termination solution, and the absorbance value is measured using a microplate analyzer at 450 nm detection wavelength with a correction wavelength of 570-630 nm.

Background：

Patients with advanced/metastatic cancer have higher levels of IL-6 in their blood.
