Testing Principle：

The double antibody sandwich ELISA is the technique employed in this kit. The high affinity conjugate plate comes pre-packaged with a specific anti-human CXCL13 capture antibody. The process involves adding the standard substance, sample to be tested, and biotin-labeled detection antibody in sequence to the HRP-plate well, followed by thorough oscillation and mixing. The incubation process takes place at room temperature for 2 hours, during which CXCL13 in the sample combines with the solid phase antibody and detection antibody. After washing to remove free and unbound ingredients, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is introduced, followed by a further wash. The next step is the addition of TMB color substrate, which incubates in the dark at room temperature for color development. The concentration of CXCL13 in the sample is directly proportional to the depth of color reaction observed. The reaction is terminated with the addition of the termination solution, and the absorbance value is measured using a microplate analyzer at a detection wavelength of 450 nm (with correction wavelength 570-630 nm).

Background：

Interleukin 4 (IL-4) is a type of cytokine which plays a key role in inducing the differentiation of young helper T cells (known as Th0 cells) into Th2 cells. When activated by IL-4, Th2 cells secrete even more of the cytokine in response.
