Testing Principle：

The double antibody sandwich ELISA kit utilizes a prepackaged high affinity conjugate plate with a specific anti-human CXCL13 Capture Antibody. First, the standard substance, sample to be tested, and biotin labeled detection antibody are added sequentially into the well of the HRP-plate. The mixture is thoroughly mixed and oscillated, and then left at room temperature for a 2-hour incubation process, allowing the CXCL13 in the sample to bind with the solid phase antibody and detection antibody. Next, thorough washing is done to remove any free and unbound components. Horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is then added and the plate is washed again. Following this, the TMB color substrate is added and the plate is incubated in darkness at room temperature, allowing for color development. The intensity of the color reaction is directly proportional to the concentration of CXCL13 in the sample. To terminate the reaction, a termination solution is added, and the absorbance value is measured at a detection wavelength of 450 nm using a microplate analyzer. It is important to note that a correction wavelength of 570-630 nm is utilized during the measurement process.

Background：

IL-17A, also known as interleukin 17A or IL-17, belongs to the IL-17 family of cytokines. It is primarily produced by helper T lymphocytes and can be stimulated by IL-23. One of its notable functions is recruiting monocytes and neutrophils to sites of inflammation by increasing the production of chemokines in tissues. This action helps to delay the onset of inflammation. In response to an extracellular pathogen invading the immune system, IL-17 acts as a pro-inflammatory cytokine and aids in destroying the pathogen's cells. It works synergistically with tumor necrosis factor and IL-1. The IL-17 family has been associated with various immune and autoimmune-related diseases, including rheumatoid arthritis, asthma, lupus, allograft rejection, anti-tumor immunity, psoriasis, and multiple sclerosis.
