Testing Principle:
The double antibody sandwich ELISA method is utilized in this kit. The conjugate plate comes prepackaged with a specific anti-human CXCL13 Capture Antibody, which exhibits high affinity. The process involves sequentially adding the standard substance, the sample to be tested, and the biotin labeled detection antibody into the HRP-plate well. After thoroughly oscillating and mixing, the sample containing CXCL13 is subjected to a 2-hour incubation at room temperature. During this incubation, the CXCL13 in the sample binds to both the solid phase antibody and the detection antibody.
To ensure accuracy, free and unbound constituents are removed via extensive washing. Subsequently, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is introduced. Following another round of washing, TMB color substrate is added and left to incubate in darkness at room temperature. The color development process occurs, and the depth of color reaction directly relates to the concentration of CXCL13 present in the sample.
To terminate the reaction, a termination solution is added. The absorbance value at a detection wavelength of 450 nm (with a correction wavelength of 570-630 nm) is measured using a microplate analyzer. This measurement provides valuable information about CXCL13 concentration.
