Testing Principle:
This ELISA kit utilizes the double antibody sandwich method, specifically the anti-human CXCL13 Capture Antibody prepackaged on a conjugate plate with high affinity. The test samples, standard substance, and biotin labeled detection antibody are sequentially added into the HRP-plate well, and allowed to incubate at room temperature for 2 hours. During this incubation process, CXCL13 in the sample binds with the solid phase antibody and detection antibody. After thorough washing to remove unbound components, horseradish peroxidase labeled streptavidin is introduced, followed by another round of washing. TMB color substrate is then added and allowed to incubate in the dark at room temperature for color development. The depth of the color reaction is directly proportional to the concentration of CXCL13 in the sample. Finally, a termination solution is added to stop the reaction, and the absorbance value is measured at 450 nm detection wavelength (with a correction wavelength of 570-630 nm) using a microplate analyzer. Overall, this kit provides a reliable and accurate method for the detection of CXCL13 levels in human samples.

TNF-α, also known as cachexin or TNFSF1A, is an adipokine that plays an important role in systemic inflammation and the stimulation of acute phase responses. It is primarily produced by activated macrophages but can also be secreted by other cells such as CD4+ lymphocytes, eosinophils, and neurons. TNF-α has a range of regulatory functions in the immune response and can also function as a potential pyrogen. Additionally, it can regulate metabolic activity in various tissues and induce local coagulation, thereby demonstrating tumor-killing activity. Inflammation in joint tissues and other tissues is also linked to the pathogenesis of TNF-α. Recently, research shows that TNF-α is involved in the pathogenesis of AIDS.
