Testing Principle：

This particular kit employs the use of double antibody sandwich ELISA. The conjugate plate is preloaded with a specific anti-human CXCL13 Capture Antibody that has high affinity. Thereafter, the standard substance, as well as the sample to be tested, is sequentially added into the HRP-plate well alongside the biotin-labelled detection antibody. The mixture is then thoroughly oscillated and thoroughly stirred, before being allowed to incubate for about two hours at ambient temperature. During the incubation, the CXCL13 in the sample combines with the detection and solid-phase antibodies. Once the incubation period has elapsed, the mixture is thoroughly washed to remove any unbound components. The next step is to add the horseradish peroxidase-labeled streptavidin-HRP (SA-HRP) before washing again. After that, TMB color substrate is added before being left to develop colour at room temperature in the dark. The depth of the colour reaction is directly related to the amount of CXCL13 that is present in the sample. The reaction is eventually terminated by the addition of termination solution before the absorbance value is measured, using a microplate analyzer, at 450 nm detection wavelength (correction wavelength 570-630 nm).

Background：
