Testing Principle：

This ELISA kit employs a double antibody sandwich technique, utilizing a specific anti-human CXCL13 Capture Antibody pre-coated onto a high affinity conjugate plate. The standard substance and the sample to be tested are added sequentially into the HRP-plate well along with a biotin-labeled detection antibody. Following full mixing and oscillation, the sample is left to incubate at room temperature for a period of two hours, during which time CXCL13 in the sample binds to the solid phase antibody and detection antibody. The excess and unbound ingredients are then thoroughly washed away, before horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is added, followed by a final washing step. The TMB color substrate is then introduced, with a subsequent incubation period in the dark at room temperature to allow color development. The depth of color reaction is directly proportional to the concentration of CXCL13 present in the sample. The reaction is terminated by the addition of termination solution, after which the absorbance value is measured at 450 nm detection wavelength (with correction wavelength 570-630 nm) using a microplate analyzer.

Background：

IL-33, a member of the IL-1 family, is secreted by various cell types like macrophages, dendritic cells, and endothelial cells. It induces the production of type II cytokines by basophils, eosinophils, and helper T cells. IL-33 triggers signaling pathways like NF-κB and MAP kinase by binding to the receptor ST2 and IL1RAP. This signaling cascade results in the secretion of type II cytokines that promote the polarization of Th2 cells. To achieve its biological function, IL-33 instigates the production of type II cytokines in vivo, which can lead to severe pathological changes in mucosal organs.
