Testing Principle：

This ELISA kit employs the double antibody sandwich method, using a specific anti-human CXCL13 Capture Antibody preloaded onto a high-affinity conjugate plate. The sample being tested, as well as a standard substance and biotin-labeled detection antibody, are subsequently added to the HRP-plate well. The sample is then incubated for two hours at room temperature, allowing it to bind with both the solid-phase antibody and the detection antibody. After thorough washing to remove any unbound components, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is added. Following another round of washing, the TMB color substrate is introduced and incubated in a dark area at room temperature to allow for color development. The intensity of the color reaction is directly proportional to the concentration of CXCL13 in the sample. Finally, the termination solution is added to halt the reaction, and the absorbance value is measured using a microplate analyzer at a detection wavelength of 450 nm (with a correction wavelength of 570-630 nm).

Background：

IL-35 is a cytokine that belongs to the IL-12 family and is mainly produced by regulatory T cells. It has been shown to play a vital role in suppressing the immune system's inflammatory response. IL-35 is made up of two independent genes, IL-12a and EBI3, which encode for the IL-12α and IL-27β chains, respectively. Although IL-35 is not consistently expressed in tissues, it can be produced by certain cells, such as monocytes, smooth muscle cells, and vascular endothelial cells, after an inflammatory stimulus. Studies in mice have shown that the absence of the IL-35 chain reduces the body's ability to suppress inflammation. Moreover, IL-35 has an inhibitory effect on specific T cell subsets, such as the pro-inflammatory Th17 cells, while promoting the proliferation of Tregs. Overall, IL-35 is an essential cytokine in controlling the immune system's activity by suppressing the inflammatory response and promoting immune tolerance.
