Testing Principle:
The double antibody sandwich ELISA kit employs a specific anti-human CXCL13 Capture Antibody that is already coated on a high affinity conjugate plate. The procedure begins by sequentially adding a standard substance, the sample to be tested, and a biotin-labeled detection antibody into the well of an HRP plate. The mixture is thoroughly mixed, allowing for a 2-hour incubation of the CXCL13 in the sample at room temperature. During this incubation process, the CXCL13 in the sample combines with the solid-phase antibody and the detection antibody. To eliminate any unbound components, the well is thoroughly washed, and horseradish peroxidase-labeled streptavidin-HRP (SA-HRP) is added. After washing again to remove excess SA-HRP, TMB color substrate is introduced and incubated in darkness at room temperature to initiate color development. The intensity of the color reaction is directly proportional to the concentration of CXCL13 in the sample. To stop the reaction, a termination solution is added, and the absorbance value is measured using a microplate analyzer at a detection wavelength of 450 nm (with a correction wavelength range of 570-630 nm).
