Testing Principle：

The double antibody sandwich ELISA kit utilizes a specific anti-human CXCL13 capture antibody that is preloaded onto a high affinity conjugate plate. The process involves the sequential addition of standard substance, sample to be tested, and biotin labeled detection antibody into the HRP-plate well. Incubation of the sample is carried out at room temperature for two hours, during which the CXCL13 in the sample combines with the solid phase antibody and detection antibody on the plate. Thorough washing is done to remove free and unbound substances before horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is added. After additional washing, TMB color substrate is added and incubation is carried out in the dark at room temperature to promote color development. The depth of color reaction is directly related to the concentration of CXCL13 in the sample. A termination solution is added to stop the reaction, and the absorbance value is measured at 450 nm detection wavelength (corrected wavelength 570-630 nm) using a microplate analyzer.

Background：

Galectin-3, a member of the lectin family, is widely expressed in various cellular compartments including the nucleus, cytoplasm, mitochondria, cell surface, and extracellular space. It exhibits an affinity for β-galactosidase and displays antimicrobial and antifungal properties. The diverse biological functions of galectin-3 encompass cell adhesion, activation, and chemotaxis, as well as cell growth, differentiation, cycle regulation, and apoptosis. Due to its multifaceted role, galectin-3 is believed to be implicated in several health conditions such as cancer, inflammatory diseases, fibrosis, heart disease, and stroke.
