Testing Principle：

The double antibody sandwich ELISA is utilized in this kit, with a specific anti-human CXCL13 Capture Antibody pre-packaged on a high-affinity conjugate plate. The sample to be tested, standard substance, and biotin-labeled detection antibody are sequentially added into the HRP-plate well, followed by thorough oscillation and mixing. The sample is then incubated at room temperature for two hours, during which the CXCL13 in the sample binds with the solid-phase antibody and detection antibody. After subsequent washing to remove free and unbound components, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is introduced. The TMB color substrate is added then placed in the dark at room temperature for color development. The intensity of the color reaction is directly proportional to the concentration of CXCL13 in the sample. To terminate the reaction, the termination solution is added, and the absorbance value is measured at 450 nm detection wavelength using a microplate analyzer, with a correction wavelength between 570-630 nm.

Background：

Brain-derived neurotrophic factor (BDNF) belongs to the neurotrophic family of growth factors and is associated with nerve growth factors. These neurotrophic factors are found in both the brain and the peripheral areas of the body. BDNF is synthesized and processed within the endoplasmic reticulum and is released by dense core vesicles. It exerts its effects on various regions of the brain, such as the hippocampus, cortex, and basal forebrain, as well as in other parts of the body including the retina, motor neurons, kidneys, saliva, and prostate. BDNF plays a crucial role in the survival of existing neurons and contributes to the growth and differentiation of new neurons and synapses in both the central and peripheral nervous systems. Changes in BDNF levels have been implicated in the development of conditions like schizophrenia, depression, eczema, epilepsy, Alzheimer's disease, obesity, aging, congenital central hyponventilation syndrome, and cognitive dysfunction after chemotherapy.
