Overview

Description

SHIP is a phosphatase that contains SH2 and regulates various cellular processes, including cell survival, growth, cell cycle arrest, and apoptosis. This hemopoietic-specific protein hydrolyzes Ins(1,3,4,5)P4 and PtdIns(3,4,5)P3. It is localized in the cytosol and possesses an SH2 domain at its N-terminus and two NPXY Shc binding motifs at its C-terminus.

Other names

The inositol polyphosphate 5 phosphatase of 145kDa, also known as SHIP-1, is an important enzyme involved in regulating cellular signaling pathways. It is encoded by the INPP5D gene and plays a key role in phosphatidylinositol signaling. The enzyme is also referred to as 4,5-trisphosphate 5-phosphatase 1 or Phosphatidylinositol-3 and has a molecular weight of 145kDa. Other aliases include hp51CN, p150Ship, and SIP-145. The protein contains an SH2 domain, which enables it to interact with other signaling proteins. Mutations in the INPP5D gene have been associated with various diseases, including cancer and autoimmune disorders. The precise function of SHIP-1 is still being investigated, but it is known to be involved in modulating immune cell activation, insulin signaling, and platelet activation.

Acts as a negative regulator of B-cell antigen receptor signaling. Mediates signaling from the FC-gamma-RIIB receptor (FCGR2B), playing a central role in terminating signal transduction from activating immune/hematopoietic cell receptor systems. Tissue specificity

Specifically expressed in immune and hematopoietic cells. Expressed in bone marrow and blood cells. Levels vary considerably within this compartment. Present in at least 74% of immature CD34+ cells, whereas within the more mature population of CD33+ cells, it is present in only 10% of cells. Present in the majority of T-cells, while it is present in a minority of B-cells (at protein level).
