Overview

Description

CD47 is an immunoglobulin-like cell surface receptor that plays a crucial role in various cellular processes. Acting as a docking protein, CD47 facilitates the translocation of PTPN6, PTPN11, and other binding partners from the cytosol to the plasma membrane. This receptor is involved in promoting the adhesion of cerebellar neurons, facilitating neurite outgrowth, and supporting glial cell attachment.
Furthermore, CD47 is believed to be an essential player in intracellular signaling during synaptogenesis and synaptic function. It is instrumental in regulating receptor tyrosine kinase-driven cellular responses triggered by cell adhesion, growth factors, or insulin. Additionally, CD47 serves as a negative regulator for phagocytosis, mast cell activation, and dendritic cell activation.
Overall, CD47's multifaceted roles make it a crucial component in various cellular functions, reinforcing its significance in cellular communication and homeostasis.

Other names

Signal regulatory protein alpha (SIRPα) is a transmembrane glycoprotein that belongs to the CD172 antigen-like family. It is also known as brain immunoglobulin-like molecule with tyrosine-based activation motifs (Bit), p84, or macrophage fusion receptor (MFR). The SIRPα molecule consists of three isoforms: alpha-1, alpha-2, and alpha-3.
SIRPα acts as an inhibitory receptor that regulates immune cell activation and function. It plays an essential role in cell-cell communication, cell adhesion, and phagocytosis, particularly in macrophages and dendritic cells. Moreover, SIRPα is involved in various processes such as inflammation, autoimmunity, and cancer.
The SIRPα protein contains three extracellular domains, including an N-terminal V-set Ig domain and two C2-set Ig domains. Additionally, it has four intracellular tyrosine-based motifs (ITIMs) and one immunoreceptor tyrosine-based inhibition motif (ITIM) that interact with protein-tyrosine phosphatases (PTPs) such as SHP-1 and SHP-2.
SIRPα expression is widespread, notably in myeloid cells but also in other tissues such as nervous system, kidney, heart, and lung. A genetic study has revealed that a single nucleotide polymorphism (SNP) on SIRPα gene is associated with increased susceptibility to several autoimmune diseases and cancer.
In conclusion, SIRPα is a crucial immunoregulatory protein that modulates immune cell activation and function. Although its role in various cellular processes is still under investigation, the emerging evidence highlights its potential as a therapeutic target for several diseases.

Specificity

The total SHPS1 levels can be detected using an SHPS1 antibody, which specifically recognizes the endogenous levels of SHPS1 protein. This antibody is valuable for studying the expression and localization of SHPS1 in various biological samples. By rearranging the original content, we can emphasize that the SHPS1 antibody is capable of detecting the entire amount of SHPS1 protein present in a given sample.

Application

The recommended dilutions for the use of this antibody in Western blotting are within the range of 1:500 to 1:3000. For immunohistochemistry, a dilution of 1:50 to 1:200 is recommended. When used for immunofluorescence, a dilution of 1:100-400 is ideal. For ELISA involving peptides, a dilution of 1:20000-1:40000 is recommended for best results. These dilution guidelines ensure the optimal performance of the antibody in various applications, allowing for accurate and reliable detection of the target protein.

Target

Background

Immunoglobulin-like cell surface receptor for CD47. Acts as docking protein and induces translocation of PTPN6, PTPN11 and other binding partners from the cytosol to the plasma membrane. Supports adhesion of cerebellar neurons, neurite outgrowth and glial cell attachment. May play a key role in intracellular signaling during synaptogenesis and in synaptic function (By similarity). Involved in the negative regulation of receptor tyrosine kinase-coupled cellular responses induced by cell adhesion, growth factors or insulin. Mediates negative regulation of phagocytosis, mast cell activation and dendritic cell activation. CD47 binding prevents maturation of immature dendritic cells and inhibits cytokine production by mature dendritic cells.

Tissue specificity

Ubiquitous. Highly expressed in brain. Detected on myeloid cells, but not T-cells. Detected at lower levels in heart, placenta, lung, testis, ovary, colon, liver, small intestine, prostate, spleen, kidney, skeletal muscle and pancreas.

Posttranslational modification

N-glycosylated.Phosphorylated on tyrosine residues in response to stimulation with EGF, growth hormone, insulin and PDGF. Dephosphorylated by PTPN11.

Celluar localization

Extracellular region or secreted;Plasma Membrane;
