Description | The proteasome is a complex proteinase found abundantly in eukaryotic cells. It has a ring-shaped 20S core structure made up of four rings of 28 non-identical subunits. These subunits are divided into two types: alpha and beta subunits of 7 each. Proteasomes are responsible for cleaving peptides via an ATP/ubiquitin-dependent process in a non-lysosomal manner. The immunoproteasome, a modified version of the proteasome, is of particular importance, and it is primarily responsible for processing class I MHC peptides. This gene belongs to the proteasome B-type family, also known as the T1B family, and encodes for the 20S core beta subunit. The gene is located in the class II region of the MHC and is primarily induced by gamma interferon. The gene product helps replace catalytic subunit 1 (proteasome beta 6 subunit) in the immunoproteasome. It is worth noting that proteolytic processing is necessary to produce a mature subunit. |
| Other names | Proteasome subunit beta type 9, also known as LMP2 or Large multifunctional peptidase 2, is a low molecular mass protein that plays a crucial role in cellular protein degradation. It is a component of the multicatalytic endopeptidase complex and belongs to the Macropain chain 7 family. Another name for this protein is MGC70470. The PSMB9 gene, also referred to as Proteasome (prosome macropain) subunit beta type 9 or proteasome (prosome, macropain) subunit, beta type, 9 (large multifunctional peptidase 2), encodes the Proteasome beta 9 subunit. This subunit contributes to the catalytic activity of the proteasome, a protein complex responsible for degrading unnecessary or damaged proteins in the cell. Additionally, this protein is associated with other names including Proteasome chain 7, Proteasome related gene 2, Proteasome subunit beta 6i, and Proteasome subunit beta type-9. It is also known as RING12 protein or Really interesting new gene 12 protein due to its intriguing biological functions. In summary, Proteasome subunit beta type 9 (LMP2) is a vital component of the proteasome complex and is involved in protein degradation processes within the cell. Its diverse nomenclature reflects its various roles and functions in cellular physiology. |

Background | The proteasome is a multicatalytic proteinase complex which is characterized by its ability to cleave peptides with Arg, Phe, Tyr, Leu, and Glu adjacent to the leaving group at neutral or slightly basic pH. The proteasome has an ATP-dependent proteolytic activity. This subunit is involved in antigen processing to generate class I binding peptides. Replacement of PSMB6 by PSMB9 increases the capacity of the immunoproteasome to cleave model peptides after hydrophobic and basic residues. |
| Tissue specificity | Highly expressed in immature dendritic cells (at protein level). |
| Posttranslational modification | Autocleaved. The resulting N-terminal Thr residue of the mature subunit is responsible for the nucleophile proteolytic activity. |
| Celluar localization | Cytosol;Extracellular region or secreted;Nucleus; |
| UniPort | P28065 |
