Description

The proteasome is a complex proteinase found abundantly in eukaryotic cells. It has a ring-shaped 20S core structure made up of four rings of 28 non-identical subunits. These subunits are divided into two types: alpha and beta subunits of 7 each. Proteasomes are responsible for cleaving peptides via an ATP/ubiquitin-dependent process in a non-lysosomal manner. The immunoproteasome, a modified version of the proteasome, is of particular importance, and it is primarily responsible for processing class I MHC peptides. This gene belongs to the proteasome B-type family, also known as the T1B family, and encodes for the 20S core beta subunit. The gene is located in the class II region of the MHC and is primarily induced by gamma interferon. The gene product helps replace catalytic subunit 1 (proteasome beta 6 subunit) in the immunoproteasome. It is worth noting that proteolytic processing is necessary to produce a mature subunit.

Background

The proteasome is a multicatalytic proteinase complex which is characterized by its ability to cleave peptides with Arg, Phe, Tyr, Leu, and Glu adjacent to the leaving group at neutral or slightly basic pH. The proteasome has an ATP-dependent proteolytic activity. This subunit is involved in antigen processing to generate class I binding peptides. Replacement of PSMB6 by PSMB9 increases the capacity of the immunoproteasome to cleave model peptides after hydrophobic and basic residues.
