Overview

Description

MMP2 is a versatile metalloproteinase that plays a vital role in various biological processes. Its functions encompass a wide range of activities, including restructuring of the blood vessels, promoting angiogenesis, facilitating tissue repair, facilitating tumor invasion, triggering inflammation, and contributing to the rupture of atherosclerotic plaques. This remarkable proteinase not only breaks down proteins in the extracellular matrix but also exhibits its effects on multiple nonmatrix proteins such as big endothelial 1 and beta-type CGRP, thereby inducing vasoconstriction. Additionally, it possesses the capability to cleave a specific bond, Gly-|-Leu, in the protein called KISS. Moreover, emerging evidence suggests that MMP2 is implicated in the pathways leading to cell death in the myocardium. The multifaceted role of MMP2 makes it a crucial player in various physiological and pathological processes.

Other names

Collagenase Type 4 alpha, also known as 72 kDa gelatinase or 72 kDa type IV collagenase, is a protein enzyme encoded by the CLG4A gene. It is also referred to as CLG 4, CLG4A, Gelatinase A, Gelatinase alpha, Gelatinase neutrophil, or Matrix metallopeptidase 2 gelatinase A 72kDa gelatinase 72kDa type IV collagenase. This enzyme plays a crucial role in the breakdown of type IV collagen, a major component of the extracellular matrix. It is classified as a matrix metalloproteinase and is commonly abbreviated as MMP-2 or MMP2. Other aliases for this protein include MMP II, MONA, Neutrophil gelatinase, PEX, and TBE-1.

Application

This antibody has a recommended working dilution range of WB 1:500-1:2000, IHC 1/200, IF/ICC 1:100-1:500 and ELISA (peptide) 1:20000-1:40000. To ensure optimal results, it is advised to dilute the antibody within this range. The antibody has been validated for these applications and dilutions, and it is expected to perform consistently within this range. It is important to note that the dilution may need to be adjusted based on the specific experiment conditions and sample type. Overall, this antibody is a reliable tool for detecting the target protein in various applications.

Immunogen

A synthesized peptide derived from human MMP2.

Stability & Storage

To preserve the quality of the product, it is recommended to store it at a temperature of -20 °C for a period of one year. It is important to avoid subjecting the product to repeated freeze/thaw cycles, as this can significantly impact its efficacy.

Storage buffer

The storage conditions for Rabbit IgG are as follows: it should be stored at -20 °C in phosphate buffered saline with a pH of 7.4. Additionally, it should be stored in a solution containing 150mM NaCl, 0.02% sodium azide, and 50% glycerol. It is important to note that the stability of the Rabbit IgG is 12 months from the date of receipt.

Target

Background

This metalloproteinase is present ubiquitously and plays a crucial role in various processes including vascular remodeling, angiogenesis, tissue healing, infiltration of tumors, inflammation, and rupture of atherosclerotic plaques. In addition to breaking down extracellular matrix proteins, it also has the ability to affect several non-matrix proteins like big endothelial 1 and beta-type CGRP, which consequently induce vasoconstriction. It also carries out the cleavage of KISS, specifically at a Gly-|-Leu bond. Furthermore, it appears to be involved in pathways related to myocardial cell death and contributes to oxidative stress in the cardiac tissue by controlling the activity of GSK3beta, as it is capable of cleaving GSK3beta in laboratory settings. Its contribution to the formation of fibrovascular tissues is closely linked with MMP14.

Tissue specificity

Produced by normal skin fibroblasts. PEX is expressed in a number of tumors including gliomas, breast and prostate.

Posttranslational modification

Phosphorylation on multiple sites modulates enzymatic activity. Phosphorylated by PKC in vitro. The propeptide is processed by MMP14 (MT-MMP1) and MMP16 (MT-MMP3). Autocatalytic cleavage in the C-terminal produces the anti-angiogenic peptide, PEX. This processing appears to be facilitated by binding integrinv/beta3.
