Overview

Description

Granzyme B (GZMB) is an essential enzyme for the destruction of target cells in cell-mediated immune responses. This enzyme performs cleavage specifically after the amino acid aspartate (Asp), and it appears to be involved in the activation cascade of caspases, which are cysteine proteases that play a crucial role in apoptosis. GZMB cleaves caspase-3, -7, -9, and -10, generating active enzymes that mediate the execution of apoptosis. It belongs to the peptidase S1 family and is classified as a member of the Granzyme subfamily. Furthermore, GZMB can be induced by staphylococcal enterotoxin A (SEA) in peripheral blood leukocytes.

Other names

C11; Cathepsin G-like 1; also known as Cathepsin G like 1 or CCPI, is a protein that belongs to the serine protease family. It is also referred to as CGL 1, CGL1, CSP B, CSPB, CTLA-1, CTLA1, CTSGL1, or Cytotoxic serine protease B. Another name for it is Cytotoxic T lymphocyte associated serine esterase 1 or Cytotoxic T lymphocyte proteinase 2. It is also known as Cytotoxic T-lymphocyte proteinase 2 or Fragmentin 2, also called Fragmentin-2. As for its code, it is often referred to as GRAB_HUMAN, Granzyme 2, or Granzyme B (granzyme 2, cytotoxic T lymphocyte associated serine esterase 1). Other aliases include Granzyme B, Granzyme-2, GranzymeB, GRB, Gzmb, Hlp, or Human lymphocyte protein. It is a protease enzyme found in lymphocytes and is responsible for various immune functions. Another name for it is Lymphocyte protease or Protease, serine, B. It is also known as SECT or T cell serine protease 1-3E. C11 is also referred to as T-cell serine protease 1-3E.
Another protein with a similar name is Cathepsin G-like 2 or CCP X, also known as CCP-X, CGL 2, CGL2, CSP C, CSP-C, CTLA1, CTSGL2, or Cytotoxic serine protease C. It is also referred to as Cytotoxic T lymphocyte associated serine esterase 1 or Cytotoxic T lymphocyte proteinase. Another name for it is Cytotoxic T-lymphocyte proteinase or EC 3.4.21.-. It is also known as GRAH_HUMAN, Granzyme H (cathepsin G-like 2, protein h-CCPX), or Granzyme H. It is denoted as GZMH or Protein h CCPX. This protein also plays a role in immune responses and is found in lymphocytes.
Please note that while these proteins have similar names and belong to the same family, they have distinct functions and characteristics. Their roles in immune responses may differ, and they may have different targets or substrates.

Application

For Western blotting, the recommended dilution range is between 1:500 to 1:3000. If you are performing an immunohistochemistry assay, the recommended dilution range is between 1:50 to 1:200. When it comes to immunofluorescence or immunocytochemistry, the recommended dilution range is between 1:100 to 1:500. Lastly, if you are performing an ELISA using a peptide, the recommended dilution range is between 1:20000 to 1:40000. Make sure to follow these recommended dilution ranges for accurate and reliable results.

Target

Background

This enzyme is necessary for target cell lysis in cell-mediated immune responses. It cleaves after Asp. Seems to be linked to an activation cascade of caspases (aspartate-specific cysteine proteases) responsible for apoptosis execution. Cleaves caspase-3, -7, -9 and 10 to give rise to active enzymes mediating apoptosis.

Tissue specificity

By staphylococcal enterotoxin A (SEA) in peripheral blood leukocytes.
