This contact-dependent signaling is known as forward signaling when initiated by the receptor and reverse signaling when initiated by the ephrin ligand. The interaction between the cognate ligand EFNA5 and its receptor EPHA7 plays a crucial role in brain development by regulating cell-cell adhesion and repulsion. It acts as a repellent for axons and guides the corticothalamic and retinal axons to their proper destinations. Additionally, it can influence brain development through a proapoptotic activity that depends on caspase (CASP3). Activation of EPHA7 can lead to phosphorylation of key components of the ERK signaling pathway, including MAP2K1, MAP2K2, MAPK1, and MAPK3.
