Overview

Description

Caveolin-1 is a protein that may function as a scaffold within caveolar membranes. It has the unique ability to directly interact with G-protein alpha subunits, actively regulating their activity. Additionally, it plays a critical role in the stimulation of T-cell receptor (TCR)-mediated T-cell activation as a costimulatory signal. Binding with DPP4 induces T-cell proliferation and NF-kappa-B activation in a T-cell receptor/CD3-dependent manner. Classified as a member of the caveolin family, caveolin-1 forms a homooligomer and interacts with a range of proteins such as GLIPR2, NOSTRIN, SNAP25 and syntaxin. Interestingly, caveolin-1 also interacts with rotavirus A NSP4, further highlighting its diverse capabilities.

Other names

Caveolin 1, also known as CAV1 or caveolae protein, is a protein found in caveolae, small invaginations of the plasma membrane. It has a molecular weight of 22 kDa and is encoded by the BSCL3 gene. In humans, it exists in two isoforms: caveolin 1 alpha and caveolin 1 beta.
Caveolin 1 is involved in various cellular processes, including cell growth inhibition and vesicular trafficking. It plays a crucial role in the formation and maintenance of caveolae structures on the cell surface. Additionally, it is implicated in several diseases, such as lipodystrophy and PPH3 (pulmonary arterial hypertension).
The caveolin 1 protein, also referred to as VIP21 or cell growth-inhibiting protein 32, is encoded by the MSTP085 gene. It is a key component of caveolae and contributes to their structural integrity. Its presence is essential for proper caveolae function and the regulation of intracellular signaling pathways.
In summary, caveolin 1 is an important protein involved in caveolae formation and cellular processes. Its different isoforms and significant functions make it a target for studying various physiological and pathological conditions.

Application

To use this antibody effectively, recommended dilutions for Western blotting range from 1:500 to 1:3000. For immunofluorescence and immunocytochemistry, optimal dilutions are between 1:100 and 1:500. ELISA utilizing the peptide will require dilutions of 1:20000 to 1:40000 for the most accurate results. It is crucial to follow the recommended dilution guidelines for each application to achieve maximum performance and accuracy.

Immunogen

A peptide that has been created by synthesizing human Caveolin-1 is currently available. This peptide is very similar to the original human Caveolin-1, and it has been found to be effective in various applications. By utilizing modern technology, researchers have been able to produce a synthesized peptide that closely matches the properties of natural Caveolin-1. This sophisticated process has yielded a valuable substance that has the potential to be useful in various fields, from medicine to biotechnology. With this synthesized peptide, researchers now have a valuable tool that can help them further their understanding of the human body and its functions.

Stability & Storage

The product should be stored at a temperature of -20 °C for a duration of one year. It is important to prevent any freeze/thaw cycles from occurring repeatedly.

Storage buffer

The storage temperature for Rabbit IgG is -20 °C, and it should be stored in phosphate buffered saline at pH 7.4, containing 150mM NaCl, 0.02% sodium azide, and 50% glycerol. It is recommended to use the product within 12 months from the date of receipt to ensure stability.

Target

Background

This protein may serve as a supportive structure within caveolar membranes and participate in various cellular processes. One of its functions is to interact with G-protein alpha subunits and modulate their activity. Additionally, it plays a crucial role in the signaling pathways that contribute to T-cell activation by providing a co-stimulatory signal necessary for T-cell receptor-mediated activation. Through its interaction with DPP4, it induces T-cell proliferation and activates NF-kappa-B in a T-cell receptor/CD3-dependent manner. It also facilitates the localization of CTNNB1 to caveolar membranes and potentially modulates signaling through the Wnt pathway. Another function of this protein is to inhibit the activation of SMAD2/3 by TGFB1. It achieves this by promoting the internalization of TGFBR1 from membrane rafts, leading to its subsequent degradation. Moreover, it is responsible for recruiting CAVIN proteins (CAVIN1/2/3/4) to the caveolae, contributing to the formation and maintenance of these membrane structures.

Tissue specificity

Skeletal muscle, liver, stomach, lung, kidney and heart (at protein level). Expressed in the brain.

Posttranslational modification

Ubiquitinated. Undergo monoubiquitination and multi- and/or polyubiquitination (PubMed:21822278). Monoubiquitination of N-terminal lysines promotes integration in a ternary complex with UBXN6 and VCP which promotes oligomeric CAV1 targeting to lysosomes for degradation (PubMed:23335559).The initiator methionine for isoform 2 is removed during or just after translation. The new N-terminal amino acid is then N-acetylated.Phosphorylated at Tyr-14 by ABL1 in response to oxidative stress.
