Description | Complement factor B, encoded by this gene, plays a crucial role in the alternative pathway of complement activation. It is found in the bloodstream as a single chain polypeptide. When the alternative pathway is activated, complement factor D cleaves it, resulting in the formation of two subunits: Ba and Bb. Bb acts as a serine protease and combines with C3b to form the alternative pathway C3 convertase. This active subunit, Bb, promotes the proliferation of preactivated B lymphocytes, while Ba restricts their proliferation. The gene is located in the major histocompatibility complex (MHC) class III region on chromosome 6, which encompasses various genes involved in regulating immune reactions. Interestingly, variations in this gene have been linked to a decreased risk of age-related macular degeneration. Notably, the polyadenylation site of this gene is positioned 421 bp away from the 5' end of the gene responsible for complement component 2. |
| Other names | The C3/C5 convertase is a key player in the complement system, which is an important part of the immune system. Complement factor B is an essential component of this convertase complex. Another important protein involved in this process is the glycine-rich beta glycoprotein, also known as PBF2. Finally, properdin factor B is another critical component of the complement system. Together, these proteins work to help the body fight off infections and other threats. |

Background | Factor B which is part of the alternate pathway of the complement system is cleaved by factor D into 2 fragments: Ba and Bb. Bb, a serine protease, then combines with complement factor 3b to generate the C3 or C5 convertase. It has also been implicated in proliferation and differentiation of preactivated B-lymphocytes, rapid spreading of peripheral blood monocytes, stimulation of lymphocyte blastogenesis and lysis of erythrocytes. Ba inhibits the proliferation of preactivated B-lymphocytes. |
