Other names

The protein p16INK4a, also known as CDKN2A, is a cell cycle inhibitor that is involved in regulating the progression of the cell cycle. It binds to and inhibits the activity of the cyclin-dependent kinase 4 (CDK4) and CDK6, which are important for cell cycle progression. In this way, p16INK4a acts as a tumor suppressor by preventing the uncontrolled cell growth that can lead to cancer.
p16INK4a is encoded by the CDKN2A gene, which is located on chromosome 9p21. This gene also encodes another protein, p14ARF (p19ARF in humans), which is involved in regulating the p53 tumor suppressor protein.
Loss of function mutations or deletions in the CDKN2A gene are associated with an increased risk of many types of cancer, including melanoma, pancreatic cancer, and others. In contrast, activation of p16INK4a expression can lead to cellular senescence and may be a potential therapeutic target for cancer treatment.

This antibody specifically recognizes and binds to the native form of p14 ARF protein present in cells. Consequently, when p14 ARF is expressed at normal levels within the cell, the p14 ARF Ab can effectively detect it. This detection method is valuable in studying the abundance and localization of p14 ARF, providing insights into its functional roles in various cellular processes.

For Western blotting applications, the recommended dilution ranges from 1:500 to 1:3000. When performing immunohistochemistry, we suggest dilutions ranging from 1:50 to 1:200. For immunofluorescence or immunocytochemistry experiments, dilute the antibody at a range of 1:100 to 1:500. For ELISA using peptides, appropriate dilutions would be 1:20000 to 1:40000. This protein possesses the remarkable ability to halt the cell cycle progression, specifically in the G1 and G2 phases. It acts as a safeguard against tumor formation by operating as a tumor suppressor. One of its important functions is to bind with MDM2, a protein responsible for shuttling between the nucleus and cytoplasm. By sequestering MDM2 within the nucleolus, it effectively impedes the oncogenic actions of MDM2. This inhibition prevents the degradation of p53, ultimately promoting increased p53-dependent transactivation and apoptosis.
Interestingly, this protein also induces cell cycle arrest in G2 and triggers apoptosis through a p53-independent mechanism. It achieves this by hindering the activation of cyclin B1/CDC2 complexes. Additionally, it interacts with other key proteins like BCL6, where it participates in downregulating BCL6-induced transcriptional repression. Moreover, this protein binds to E2F1 and MYC, restricting their transcriptional activation abilities, while leaving their repressive actions unaffected.
Furthermore, it forms a complex with TOP1/TOPOI, stimulating its activity. This complex binds to promoters of rRNA genes, indicating a potential role in rRNA transcription and/or maturation. It may also impact rRNA processing by interacting with NPM1/B23, promoting its polyubiquitination and subsequent degradation. Along similar lines, it interacts with COMMD1 and facilitates its 'Lys63'-linked polyubiquitination. Another notable interaction occurs with UBE2I/UBC9, enhancing the process of sumoylation for various binding partners, including MDM2 and E2F1. Interestingly, it also binds with HUWE1 and curtails its ubiquitin ligase activity.
Given its diverse array of interactions and functions, this protein likely plays a significant role in balancing cell proliferation and apoptosis during mammary gland development. Additionally, it's worth mentioning that a specific isoform of this protein, known as smARF, might be involved in regulating autophagy and caspase-independent cell death. Notably, the shorter-lived mitochondrial isoform finds stabilization through C1QBP.

Posttranslational modification

Ubiquitinated in normal cells by TRIP12 via the ubiquitin fusion degradation (UFD) pathway, a process that mediates ubiquitination at the N-terminus, regardeless of the absence of lysine residues. Ubiquitination leads to its proteasomal degradation. In cancer cells, however, TRIP12 is located in a different cell compartment, preventing ubiquitination and degradation.
