Other namesThe telomeric DNA binding protein, also known as telomeric DNA-binding protein or TTAGGG repeat binding factor 2, plays a crucial role in maintaining the integrity of telomeres, the protective caps at the ends of chromosomes. Its official gene symbol is TERF2, although it is also referred to as TRF2, telomeric repeat-binding factor 2, or TTAGGG repeat-binding factor 2.
This protein binds specifically to the TTAGGG repeat sequence found at the ends of chromosomes and helps to prevent the loss of genetic information during cell division. Additionally, TERF2 has been shown to help regulate the structure and length of telomeres, which is important for preventing the premature aging of cells.
Research has also suggested that TERF2 may have a role in the development and progression of cancer, as it is often found to be overexpressed in tumor cells. This knowledge could potentially lead to the development of new therapies targeting TERF2 in certain cancers.
Overall, the telomeric DNA binding protein is an essential component of the telomere complex and plays a crucial role in maintaining genomic stability and preventing age-related diseases.
SourceRabbit
SpecificityThe antibody TRF2 (Phospho-Thr188) is specifically designed to detect the presence of phosphorylated Thr188 in endogenous levels of TRF2. This means that it is capable of identifying TRF2 only when it is in a phosphorylated state at Thr188.

purificationThe purified rabbit serum was used to extract the antibody through a process of affinity purification. This involved sequential chromatography on phospho- and non-phospho-peptide affinity columns to ensure the highest level of similarity between the extracted antibody and the original serum.

Stability & StorageTo ensure long-term storage, it is recommended to keep the product at a temperature of -20 °C for up to one year. It is important to avoid subjecting the product to repeated freeze/thaw cycles, as this can affect its stability. Take note of these storage guidelines to maintain the product's quality and effectiveness.
Storage bufferRabbit IgG in phosphate buffered saline , pH 7.4, 150mM NaCl, 0.02% sodium azide and 50% glycerol.Store at -20 °C.Stable for 12 months from date of receipt.

Target

Background

Binds the telomeric double-stranded 5'-TTAGGG-3' repeat and plays a central role in telomere maintenance and protection against end-to-end fusion of chromosomes. In addition to its telomeric DNA-binding role, required to recruit a number of factors and enzymes required for telomere protection, including the shelterin complex, TERF2IP/RAP1 and DCLRE1B/Apollo. Component of the shelterin complex (telosome) that is involved in the regulation of telomere length and protection. Shelterin associates with arrays of double-stranded 5'-TTAGGG-3' repeats added by telomerase and protects chromosome ends; without its protective activity, telomeres are no longer hidden from the DNA damage surveillance and chromosome ends are inappropriately processed by DNA repair pathways. Together with DCLRE1B/Apollo, plays a key role in telomeric loop (T loop) formation by generating 3' single-stranded overhang at the leading end telomeres: T loops have been proposed to protect chromosome ends from degradation and repair. Required both to recruit DCLRE1B/Apollo to telomeres and activate the exonuclease activity of DCLRE1B/Apollo. Preferentially binds to positive supercoiled DNA. Together with DCLRE1B/Apollo, required to control the amount of DNA topoisomerase (TOP1, TOP2A and TOP2B) needed for telomere replication during fork passage and prevent aberrant telomere topology. Recruits TERF2IP/RAP1 to telomeres, thereby participating in to repressing homology-directed repair (HDR), which can affect telomere length.

Tissue specificityUbiquitous. Highly expressed in spleen, thymus, prostate, uterus, testis, small intestine, colon and peripheral blood leukocytes.
Posttranslational modificationPhosphorylated upon DNA damage, most probably by ATM. Phosphorylated TERF2 is not bound to telomeric DNA, and rapidly localizes to damage sites.Methylated by PRMT1 at multiple arginines within the N-terminal Arg-rich region. Methylation may control association with telomeres.
