Overview | |
Other names | The Nanog homeobox gene encodes for the Homeobox protein NANOG, also known as hNanog. This protein plays a crucial role in maintaining the pluripotency of embryonic stem cells, making it a key gene in the field of developmental biology. The transcription factor encoded by Nanog is involved in sustaining the undifferentiated state of these cells, and has been shown to be necessary for the generation of embryonic stem cells in mammals. Mutations in this gene have been linked to abnormal cell differentiation and tumorigenesis. Therefore, understanding the function and regulation of Nanog is critical for advancing the field of regenerative medicine and for developing targeted cancer therapies. |

Background | This particular gene contains the instructions for creating a protein that acts as a transcription factor, binding to DNA and regulating gene activity. Its role is crucial in the proliferation, renewal, and maintenance of embryonic stem (ES) cells, ensuring their pluripotency, or ability to differentiate into various cell types. Interestingly, this protein has the ability to impede ES cell differentiation, preventing them from specializing into specific cell types. Moreover, during the process of cell differentiation, it can also repress its own production. Notably, different forms of this gene can give rise to distinct isoforms of the protein, resulting in functional diversity. |

Data Examples

The analysis involved the use of NANOG antibody (abs116349) at a dilution of 1:1000 to examine extracts from different cell lines using the Western blot technique. A secondary antibody, specifically HRP Goat Anti-Rabbit IgG (H+L) at a dilution of 1:10000, was used. Each lane of the gel contained 25ug of lysates/proteins. To block non-specific binding, a blocking buffer comprising of 3% nonfat dry milk in TBST was utilized. Detection was achieved using the ECL Basic Kit, with an exposure time of only 1 second.
