Activity

Spectrophotometric analysis at 340nm was utilized to assay for Malic Enzyme activity, in line with the methodology outlined in Mandela and Sauer's work from 1975. The standard reaction mixture was composed of Tris-HCl at a concentration of 50mM, manganese chloride (MnCl2) at 3mM, malate set at 5mM, NADP+ at 0.12mM, and fumarate at 2.5mM. The measurement was conducted using a Beckman spectrophotometer. The calculated Km value is approximately 1.5mM, with an error margin of ±0.6mM.

Reconstitution

To ensure the proper distribution of the contents, it is recommended to briefly centrifuge the vial before opening. For reconstitution, use sterile distilled water or an aqueous buffer solution that contains 0.1% BSA and adjust the concentration to 0.1-1.0 mg/mL. Stock solutions should be divided into working aliquots and stored at a temperature no higher than -20 °C. Any further dilutions required should be made in appropriate buffered solutions.

Stability & Storage

This lyophilized preparation is stable at 2-8 °C, but should be kept at -20 °C for long term storage, preferably desiccated. Upon reconstitution, the preparation is stable for up to one week at 2-8 °C. For maximal stability, apportion the reconstituted preparation into working aliquots and store at -20 °C to -70 °C. Avoid repeated freeze/thaw cycles.
