Background

The lactose operon repressor (LacI) is an essential protein that consists of a HTH lacI-type DNA-binding domain and operates as a homotetramer. Being a repressor of the lactose operon, LacI acts as an inducer by binding allolactose. If you remove residues 1-59, it leads to the loss of DNA-binding activity. However, the structure of the tetramer remains intact, and inducer-binding activity is still present. On the other hand, deleting residues 340-360 leads to the loss of tetramer formation. Still, dimer formation, inducer-binding activity, and DNA-binding activity remain present.
