Testing Principle：

The double antibody sandwich ELISA method is employed in this kit. A conjugate plate with a high affinity for specific anti-human CXCL13 Capture Antibody is provided. The process involves the successive addition of standard substance, the sample to be tested, and a biotin-labeled detection antibody onto the HRP-plate well. Following thorough oscillation and mixing, the sample is incubated at room temperature for 2 hours, allowing the CXCL13 in the sample to bind with the solid phase antibody and detection antibody. To remove any free and unbound components, the well is thoroughly washed. Next, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is introduced. Another round of washing is performed before the addition of TMB color substrate. This mixture is incubated in the dark at room temperature to initiate color development. The intensity of the color reaction directly correlates with the concentration of CXCL13 in the sample. To halt the reaction, a termination solution is added, and the absorbance value is measured at a detection wavelength of 450 nm (with a correction wavelength of 570-630 nm) using a microplate analyzer.

Background：

IL-6, a cytokine with both pro-inflammatory and anti-inflammatory properties, is composed of 187 amino acids in adult mice. Interestingly, when comparing its amino acid sequence, mouse IL-6 shows a 39% and 85% sequence homology with white IL-6 in humans and rats, respectively.
IL-6 is a cytokine that is produced by T cells and macrophages to promote the immune response during infections and after injuries. It is especially useful in cases of inflammation caused by burns or other kinds of tissue damage. Research has revealed that IL-6 is a necessary component in combating Streptococcus pneumoniae infection in mice, indicating that it plays a crucial role in infection resistance.
IL-6 promotes inflammatory and autoimmune disease progression in many diseases such as diabetes, atherosclerosis, depression, Alzheimer's disease, systemic lupus erythematosus, multiple myeloma, prostate cancer, Behcet's disease, and rheumatoid arthritis. Patients with advanced/metastatic cancer have higher levels of IL-6 in their blood.
