Testing Principle：

The double antibody sandwich ELISA is utilized in this kit. The high affinity conjugate plate already contains the specific anti-human CXCL13 Capture Antibody. Firstly, the standard substance, as well as the sample to be tested, are added into the HRP-plate well. Then, the biotin labeled detection antibody is added and mixed thoroughly. The sample, containing CXCL13, is left to incubate at room temperature for 2 hours, which allows for the combination of CXCL13 with the solid phase antibody and detection antibody. To remove any unbound ingredients, a thorough washing is done. Next, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is added. Following another washing step, TMB color substrate is added and incubated in the dark at room temperature to initiate color development. The intensity of the color reaction is directly proportional to the concentration of CXCL13 present in the sample. To stop the reaction, a termination solution is added. Finally, the absorbance value is measured at a detection wavelength of 450 nm (with a correction wavelength of 570-630 nm) using a microplate analyzer. Background：

IL-5 is a Th2 cytokine that belongs to the hematopoietic family of proteins. It consists of 115 amino acids and unlike other cytokines such as IL-3 and GM-CSF, IL-5 exists in its active form as a homodimer. Th2 cells and mast cells are sources of IL-5 secretion. As a mediator of Th2 cell activity, IL-5 is biologically active on B cells and other cells. Binding of IL-5 to its receptor results in the growth of B cells and increased secretion of immunoglobulin. Additionally, IL-5 plays a critical role in the activation of eosinophils. IL-5 has been linked to various diseases including asthma and allergic rhinitis. An increase in IL-5 has been observed in circulatory and tracheal tissues while sputum eosinophils decrease.
