Testing Principle

The Rat TNF-α ELISA Kit follows a sandwich enzyme immunoassay technique to obtain quantitative results. The process involves using a high affinity plate that is pre-coated with a specific anti-rat TNF-α antibody. Subsequently, biotinylated detection antibodies, along with standards and test samples, are added to the wells of the enzyme-labeled plate. The TNF-α present in the samples combines with the solid-phase antibodies and detection antibodies, leading to the formation of immune complexes. The unbound material is washed, and horseradish peroxidase-labeled streptavidin (HRP) is introduced. After washing, a chromogenic substrate is added, and the color develops in the dark. The reaction is stopped by adding a stop solution, and the absorbance is measured at a wavelength of 450 nm (reference correction wavelength of 540 nm or 570 nm)..In summary, this kit effectively quantifies the levels of TNF-α in rat samples through an easy and efficient process.

Background: TNF-α 

Tumor necrosis factor alpha, or TNF-α, is a well-known member of the tumor necrosis factor superfamily. As a multifunctional factor, TNF-α plays a crucial role in modulating inflammation, immune system development, programmed cell death, and lipid metabolism. However, it is also implicated in various pathological conditions such as asthma, Crohn's disease, rheumatoid arthritis, neuropathic pain, obesity, type 2 diabetes, septic shock, autoimmune disorders, and cancer. Overall, TNF-α is an important molecular mediator that controls different biological processes and can be both beneficial and detrimental depending on the context.


TNF-α, a 26 kDa transmembrane protein, is present in humans. It is composed of different regions including an intracellular domain of 35 amino acids, a transmembrane segment of 21 amino acids, and an extracellular domain (ECD) of 177 amino acids. The ECD of human TNF-α shares a high amino acid sequence similarity of 97% with rhesus monkey, while it shows a range of 71-92% homology with various species such as cow, dog, cotton mouse, horse, cat, mouse, pig, and rat. Numerous cell types, including immune cells, epithelial cells, endothelial cells, and tumor cells, are capable of expressing TNF-α. The biological functions of TNF-α include the induction of targeted cell lysis in tumors and virus-infected cells. Additionally, TNF-α can bind to soluble TNF RI, leading to the generation of transmembrane signals in downstream cells. The release of active TNF-α cytokine occurs through the action of TACE/ADAM17, which promotes the shedding of cell membranes containing TNF-α. The resulting active TNF-α cytokine exists as a trimer and possesses an extracellular soluble structure with a molecular weight of 55 kDa.


Rat TNF-α ELISA Kit has two receptors,which are TNF RI and TNF RII.The molecular weight of TNF RI is 55-60 kDa and it is widely expressed.TNF RII has a molecular weight of 78-80 kDa and is just expressed in hematopoietic cells. Both of them are expressed as homologous trimer. TNF-α promotes NFκB activation with a similar binding affinity to TNF RI and TNF RII. Both the two soluble receptors are released into human serum and urine and they can neutralize TNF-α activity. However, only TNF RI has a cell death domain that can induce apoptosis.
