Testing Principle：

This ELISA kit employs the double antibody sandwich method and specifically utilizes the anti-human CXCL13 Capture Antibody that is pre-coated on a conjugate plate with high affinity. The process involves the sequential addition of the standard substance, the sample being tested, and the biotin labeled detection antibody onto the HRP-plate well. After thorough oscillation and mixing, the CXCL13 in the sample is incubated at room temperature for two hours, allowing its binding with both solid phase and detection antibodies. The unbound ingredients are then carefully washed away, following which horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is introduced into the mixture. After another round of washing, the TMB color substrate is added and kept in the dark at room temperature for color development. The depth of color reaction is directly proportional to the concentration of CXCL13 in the sample. To halt the reaction, a termination solution is added, and the absorbance value is then measured at a detection wavelength of 450 nm (with a correction wavelength of 570-630 nm) using a microplate analyzer.

Background：

B7-H1, also referred to as PD-L1 and CD274, is a membrane-bound protein weighing 40 kDa. Its function in regulating the immune system is crucial during pregnancy, organ transplantation, autoimmune disorders, hepatitis, and some other conditions. When B7-H1 binds to its receptor, it transmits inhibitory signals, which cause the suppression of CD8+ T cell proliferation in lymph nodes. B7-H1 upregulation can lead to tumor escape and metastasis. Research on renal cell carcinoma patients has shown a 4.5-fold increase in the risk of tumor invasion and death in individuals with high expression of B7-H1. B7-H1 also functions as a positive costimulatory molecule in the case of intracellular infections. In children with systemic lupus erythematosus, B7-H1 levels have been found to be altered.
