Testing Principle：

The basis of this kit is the double antibody sandwich ELISA method, which is highly effective in detecting human CXCL13. The capture antibody used in the kit is specific to anti-human CXCL13, already prepackaged on a high affinity conjugate plate. To conduct the test, the standard substance, the sample to be tested, and biotin labeled detection antibody were sequentially added to the HRP-plate well. The sample was left to incubate at room temperature for 2 hours, which allowed the CXCL13 present in the sample to combine with the solid phase and detection antibodies. Thorough washing removed any free and unbound ingredients, following which horseradish peroxidase labeled streptavidin-HRP (SA-HRP) was added. After a further washing, TMB color substrate was added and allowed to incubate in the dark at room temperature for color development. The amount of color developed was directly proportional to the CXCL13 concentration in the sample. Finally, the reaction was stopped by adding the termination solution, and the absorbance value was measured using a microplate analyzer at the 450 nm detection wavelength (with a correction wavelength range from 570-630 nm).

To further aid in the detection process, we have included SA-HRP, which helps in the visualization of the CXCL13 test results. For efficient and secure handling, a Seal Plate Film has been included.

Background：

IL-6, also known as interleukin 6, serves as a cytokine with dual properties of both pro-inflammatory and anti-inflammatory effects. It is produced by T cells and macrophages in humans, with its genetic code encoded by the IL-6 gene. The secretion of IL-6 occurs during infection and following injury, particularly in cases of inflammation caused by tissue damage or burns. Research indicates that IL-6 plays a crucial role in the immune response against Streptococcus pneumoniae infection, emphasizing its significance in enhancing resistance to infections.
IL-6 promotes inflammatory and autoimmune disease progression in many diseases such as diabetes, atherosclerosis, depression, Alzheimer's disease, systemic lupus erythematosus, multiple myeloma, prostate cancer, Behcet's disease, and rheumatoid arthritis.  Patients with advanced/metastatic cancer have higher levels of IL-6 in their blood.
