Testing Principle：

This ELISA kit is designed for the detection of human CXCL13 using a double antibody sandwich method. The high affinity conjugate plate has a specific anti-human CXCL13 Capture Antibody prepackaged for easy use. To perform the analysis, standard substance, test samples, and biotin labeled detection antibody are added successively to the HRP-plate well. After an incubation process of 2 hours at room temperature, the CXCL13 in the sample is combined with the solid-phase antibody and detection antibody. Free and unbound ingredients are removed by thorough washing, followed by the addition of horseradish peroxidase labeled streptavidin-HRP (SA-HRP). After another wash, TMB color substrate is added to develop the color reaction, and the depth of the reaction correlates positively with the concentration of CXCL13 in the sample. The reaction is stopped by adding the termination solution, and the absorbance value is measured at 450 nm detection wavelength using a microplate analyzer, with correction wavelength between 570-630 nm. This kit is an easy-to-use and reliable tool for the detection of human CXCL13.

Background：

IgM, also known as Immunoglobulin M, holds the title of being the largest antibody found in the human system. This antibody is primarily produced by B cells and is the first one to be generated in response to antigen stimulation. Specifically, it is produced by plasma cells residing in the spleen, which happens to be the main site for IgM production. While IgM can primarily be found in the serum, it also serves as a significant secretory immunoglobulin due to the J chain it possesses. During the early stages of infection, IgM makes its appearance and continues to be produced in limited quantities upon reinfection. Due to its inability to cross the human placenta, IgM antibodies play a valuable role in diagnosing infectious diseases. Detection of IgM in a patient's serum indicates a recent infection, while its presence in a newborn's serum suggests an intrauterine infection. By utilizing these two distinctive characteristics, IgM contributes greatly to the accurate diagnosis of various infectious diseases.
