Description | A20 is a protein that is induced by cytokines and plays a crucial role in inhibiting apoptosis while promoting NF-κB activation. It was first discovered in human umbilical vein endothelial cells as a TNF-α-inducible primary response gene, and its sequence shows that it contains seven Cys2/Cys2-zinc finger motifs. Constitutive expression of A20 is observed in lymphoid tissues, but stressful stimuli such as TNF-α, IL-1, phorbol esters, and LPS transiently induce its expression. By interfering with TRAF family members and interacting with the NF-κB inhibiting protein ABIN, A20 is able to confer resistance to apoptosis and NF-κB activation. Additionally, A20 has site-specific ubiquitin modifying activity that contributes to its biological functions. Its amino-terminus contains DUB activity for Lys63 branches, while the carboxyl-terminus contains E3 activity for Lys48 branches, leading to the degradation of TRAF6 and RIP substrates. Overall, A20 plays a critical role in regulating the immune response and inflammation. |
