Description

A likely transcription factor, TFEB, is known to bind to the USF/MLTF site and is believed to have affinity for the E-box sequences found in the enhancer of the heavy-chain immunoglobulin. Making use of this information, it is possible to generate alternative content that maintains the essence of the original text.

Other namesTFEB (Transcription factor EB), also known as Alpha TFEB or bHLHe35, is a Class E basic helix-loop-helix protein 35 that plays a crucial role in T cell transcription. It is an important transcription factor involved in regulating gene expression. TFEB is often identified as TCFEB or TFEB_HUMAN in scientific literature. Its function is to bind to DNA and control the activation or suppression of genes, thereby influencing cellular processes and functions. TFEB is responsible for coordinating the expression of genes involved in autophagy, lysosomal biogenesis, and lipid metabolism. This transcription factor's role in cellular homeostasis and response to stress has made it an intriguing focus for research in various fields, including cancer, neurodegenerative diseases, and metabolic disorders. Understanding the functions and regulation of TFEB could open up new avenues for therapeutic interventions targeting these conditions.
SourceRabbit
SpecificityThe detection of endogenous levels of total TFEB is possible with the TFEB Antibody. This antibody is specifically designed to recognize and bind to TFEB proteins present within cells. The reliable detection of TFEB can be attributed to the high specificity and sensitivity of this antibody. Consequently, researchers can confidently analyze and study the total TFEB levels in various biological samples.

ApplicationFor western blotting, the recommended dilution range is 1:500-1:2000, while for immunohistochemistry, it is suggested to use a dilution range of 1:50-1:200. For ELISA using peptide antigens, it is advisable to use a dilution range of 1:20000-1:40000. These dilution ranges have been determined to yield optimal results in each respective method and are therefore recommended for use.

Target

Background

Transcription factor that specifically recognizes and binds E-box sequences (5'-CANNTG-3'). Efficient DNA-binding requires dimerization with itself or with another MiT/TFE family member such as TFE3 or MITF. In association with TFE3, activates the expression of CD40L in T-cells, thereby playing a role in T-cell-dependent antibody responses in activated CD4+ T-cells and thymus-dependent humoral immunity. Specifically recognizes and binds the CLEAR-box sequence (5'-GTCACGTGAC-3') present in the regulatory region of many lysosomal genes, leading to activate their expression. It thereby plays a central role in expression of lysosomal genes. Acts as a positive regulator of autophagy by promoting expression of genes involved in autophagy. Specifically recognizes the gamma-E3 box, a subset of E-boxes, present in the heavy-chain immunoglobulin enhancer. Plays a role in the signal transduction processes required for normal vascularization of the placenta.

Posttranslational modificationSumoylated; does not affect dimerization with MITF.
