Description

On chromosome 5q13, there exists a 500 kb inverted duplication which encompasses this particular gene. Within this duplicated region, there are multiple genes and repetitive elements that increase the likelihood of rearrangements and deletions. The intricate and repetitive nature of the sequence has posed challenges in deciphering the organization of this genomic region. Specifically, this gene is located within the telomeric copy of the duplication. In some cases of spinal muscular atrophy (SMA) patients, the deletion of this gene coincides with the deletion of the neighboring SMN1 gene; however, it remains uncertain whether the absence of this gene contributes to the SMA phenotype. The encoded protein of this gene is the 44 kDa subunit of RNA polymerase II transcription initiation factor IIH, which plays a role in basal transcription and nucleotide excision repair. Although transcript variants of this gene have been identified, their complete structure is yet to be determined. Interestingly, a second copy of this gene has been reported in the literature, located within the centromeric copy of the duplication. Notably, there are contradicting reports stating a difference of either two or four base pairs between the two copies. It is important to note that there is currently no available sequence data for the centromeric copy of the gene.
