Description

TDGF-1, also known as Cripto, is a member of the EGF-CFC family of proteins that plays a significant role in embryogenesis and oncogenesis. This protein contains a conserved cysteine-rich domain (CFC motif) and a hydrophobic carboxy-terminal tail that facilitates its attachment to the extracellular plasma membrane via a GPI moiety.
The O-linked fucosylation site within the EGF-like motif is necessary for Cripto to function as a coreceptor for TGF-β-related ligands such as Nodal and Vg1/GDF1. Besides, Cripto also has paracrine activity and acts as a co-ligand for Nodal.
Cripto is highly expressed in early embryos and embryonic stem cells, where it plays significant roles in cardiomyocytic differentiation and negatively regulates neurogenesis. The protein positively regulates stem cell self-renewal and pluripotency, and its transient activation is essential for maintaining these properties in embryonic and adult-derived stem cells.
Cripto is a potent oncogene that promotes tumorigenesis by stimulating various activities such as proliferation, cell motility, invasion, angiogenesis, and epithelial-mesenchymal transition (EMT). It is highly expressed in a broad range of tumors, highlighting its importance as a therapeutic target in cancer.
