Overview

Description

p53 is directly responsible for inducing the expression of a multi-pass membrane protein called STEAP3. This protein plays a pivotal role in the facilitation of exosome secretion, particularly in the DNA damage-induced exosomal secretory pathway that relies on p53. STEAP3 primarily localizes to vesicular-like structures surrounding the nucleus and the plasma membrane. It exhibits high expression levels in hematopoietic tissues, where it localizes to the specialized endosomes.
In erythroid cells, STEAP3 acts as an endosomal ferrireductase, crucial for efficient uptake of iron through transferrin. It assists in maintaining iron homeostasis in these cells by reducing Fe(3+) to Fe(2+). Additionally, STEAP3 has the ability to reduce Cu(2+) to Cu(1+), indicating its involvement in copper homeostasis. This reduction process relies on NAD(+) as an acceptor.
To recap, STEAP3, a multi-pass membrane protein induced directly by p53, facilitates exosome secretion. Its localization varies between vesicular-like structures surrounding the nucleus and the plasma membrane. It is highly expressed in hematopoietic tissues, where it localizes to specialized endosomes. Functionally, it acts as an endosomal ferrireductase, aiding in transferrin-dependent iron uptake in erythroid cells and participating in erythroid iron and copper homeostasis by reducing Fe(3+) to Fe(2+) and Cu(2+) to Cu(1+), respectively. This reduction process utilizes NAD(+) as an acceptor.

Other names

Dudlin 2, also known as Dudlin2 or Dudulin 2, is a protein encoded by the gene 1010001D01Rik. It is also referred to as Dudulin-2 or Dudulin2, as well as hpHyde, hTSAP6, or Tumor suppressor activated pathway protein 6. Another name for this protein is Metalloreductase STEAP3 or STEAP family member 3.
This protein, with the alternative names MGC93147 or pHyde, is classified as Six transmembrane epithelial antigen of prostate 3 or Six transmembrane prostate protein 3. It plays a role as a metalloreductase and is involved in the tumor suppression pathway.
The abbreviation STEA3 or STEAP 3 is often used to refer to this protein, along with STMP 3 or TSAP 6. The full name Tumor suppressor-activated pathway protein 6 is also associated with this protein.
Overall, Dudlin 2, or STEAP3, is an important protein in various biological processes and is a potential target for therapeutic interventions.

Application

The recommended dilutions for Western blotting range from 1:500 to 1:2000, while for immunofluorescence and immunocytochemistry, the suggested dilutions are between 1:100 and 1:500. In ELISA assays using a peptide substrate, a dilution of 1:20000 to 1:40000 is commonly used. Utilizing different dilution ranges, these techniques are employed to detect and analyze specific proteins in various biological samples. It is essential to optimize the dilutions for each assay to achieve accurate and reliable results.

Immunogen

A synthesized peptide derived from human STEA3.

Stability & Storage

To maintain the quality of the product, it is recommended to store it at a temperature of -20°C for a period of one year. It is important to note that exposing the product to repeated freeze and thaw cycles may lead to a deterioration in the product's quality. Therefore, it is best to avoid such cycles and maintain a stable storage temperature throughout the product's lifespan.

Storage buffer

The storage conditions for Rabbit IgG are as follows: it should be stored at -20°C in phosphate buffered saline (pH 7.4) containing 150mM NaCl, 0.02% sodium azide, and 50% glycerol. It is important to note that the stability of this product is guaranteed for a period of 12 months from the date of receipt.

Target

Background

Endosomal ferrireductase required for efficient transferrin-dependent iron uptake in erythroid cells. Participates in erythroid iron homeostasis by reducing Fe3+ to Fe2+. Can also reduce of Cu2+ to Cu1+, suggesting that it participates in copper homeostasis. Uses NADP+ as acceptor. May play a role downstream of p53/TP53 to interface apoptosis and cell cycle progression. Indirectly involved in exosome secretion by facilitating the secretion of proteins such as TCTP.

Tissue specificity

Expressed in adult bone marrow, placenta, liver, skeletal muscle and pancreas. Down-regulated in hepatocellular carcinoma.

Posttranslational modification

Proteolytically cleaved by RHBDL4/RHBDD1. RHBDL4/RHBDD1-induced cleavage occurs at multiple sites in a glycosylation-independent manner.Glycosylated.
