Description

S100 proteins, despite their simple structure and small size, play a crucial role in regulating several cellular processes. These proteins, which range from 8-12 kDa, are involved in cell growth, cell motility, cell cycle progression, transcription, and differentiation. Currently, there are 25 known members of the S100 protein family, including S100A1-S100A18, trichohyalin, filaggrin, repetin, S100P, and S100Z, making it the largest group within the calcium-binding protein family known as EF-hand. Interestingly, 14 of these S100 genes are located on human chromosome 1q21, an area known for genomic instability. Research studies have shown a significant correlation between abnormal S100 protein expression and the progression of cancer.
S100 proteins primarily function in immune responses across various tissues, but they also play a role in neuronal development. Each S100 monomer contains two EF-hand motifs and has the capability to bind up to two calcium ions, or sometimes other divalent cations. Structurally, S100 proteins form antiparallel homo- or heterodimers and rely on the presence of calcium, although some interactions can occur in a calcium-independent manner. Despite their similarities, individual S100 members have distinct tissue distribution, cellular localization, and unique protein binding partners, indicating their specific roles in various signaling pathways. Moreover, some S100 proteins function not only intracellularly but also act as receptors for extracellular ligands or are secreted and exhibit cytokine-like activities.
One prominent member of the S100 protein family is S100B, which is abundantly expressed in astrocytes and serves as a marker for these cells in studies of the central nervous system. S100B is also found in immature and mature myelinating oligodendrocytes that express chondroitin sulfate proteoglycan (NG2).
