Description

Intracellular protein degradation is primarily carried out by the 20S proteasome, which is a crucial proteolytic enzyme complex. This complex is composed of four stacked rings, with the outer layers consisting of α subunits (PSMAs) and the inner layers consisting of β subunits. The catalytic sites are located on the β rings, while the α subunits play important roles in assembly and binding of regulatory proteins. Mammals have been found to possess seven different α and ten different β proteasome genes. Furthermore, there are three major protein complexes, namely PA700, PA28, and PA200, that act as activators for the 20S proteasome.
PA700 has a strong affinity for polyubiquitin and forms a complex with the 20S proteasome, resulting in the formation of the 26S proteasome. This complex is primarily responsible for the degradation of poly-ubiquitinated proteins. The proteasome exhibits a wide range of substrates including cell cycle regulators, signaling molecules, tumor suppressors, and transcription factors. By regulating the degradation of these intracellular proteins, the proteasome plays critical roles in various processes such as cell cycle regulation, cancer development, immune responses, protein folding, and disease progression.

Other namesThe proteasome subunit alpha type 6 is also known as IOTA, PROS-27, or p27K. It is a 27 kDa prosomal protein that belongs to the multicatalytic endopeptidase complex iota chain. Other aliases for this protein include Macropain iota chain, Proteasome subunit alpha type-6, and PSA6_HUMAN. The macropain subunit iota is encoded by genes MGC22756, MGC2333, and MGC23846. This protein plays a crucial role in cellular protein degradation and turnover. Its expression has been linked to various physiological processes, including cell cycle regulation, apoptosis, and immune response. Overall, the proteasome subunit alpha type 6 is an essential component of the proteasome machinery, whose dysfunction has been implicated in a wide range of human diseases, including cancer, neurodegenerative disorders, and viral infections.

Target

Background

Component of the 20S core proteasome complex involved in the proteolytic degradation of most intracellular proteins. This complex plays numerous essential roles within the cell by associating with different regulatory particles. Associated with two 19S regulatory particles, forms the 26S proteasome and thus participates in the ATP-dependent degradation of ubiquitinated proteins. The 26S proteasome plays a key role in the maintenance of protein homeostasis by removing misfolded or damaged proteins that could impair cellular functions, and by removing proteins whose functions are no longer required. Associated with the PA200 or PA28, the 20S proteasome mediates ubiquitin-independent protein degradation. This type of proteolysis is required in several pathways including spermatogenesis (20S-PA200 complex) or generation of a subset of MHC class I-presented antigenic peptides (20S-PA28 complex).
