Description

cAMP, a crucial signaling molecule involved in numerous cellular functions, plays a key role in activating the cAMP-dependent protein kinase. This kinase acts by phosphorylating various target proteins to transmit the signal. Initially, the inactive holoenzyme consists of a tetramer that comprises two regulatory and two catalytic subunits. Upon cAMP stimulation, the holoenzyme dissociates, resulting in a dimer of regulatory subunits bound to four cAMP molecules and two monomeric catalytic subunits. In humans, there are four distinct regulatory subunits and three catalytic subunits identified. The gene responsible for encoding the protein discussed belongs to the Ser/Thr protein kinase family and encodes a catalytic subunit of cAMP-dependent protein kinase. Notably, multiple alternatively spliced transcript variants have been observed, giving rise to different isoforms.

Other namesThe beta subunit isoform 4ab of cAMP-dependent protein kinase catalytic is also known as PKA C-beta or PKACB. It is a protein kinase that plays a crucial role in various cellular functions. This kinase is encoded by the Prkacb gene and is responsible for the transfer of phosphate groups from ATP to target proteins, leading to their activation or inactivation. Moreover, it is involved in the regulation of glycogen, sugar, and lipid metabolism. The cAMP-dependent protein kinase catalytic subunit beta is highly similar to the protein kinase cAMP dependent catalytic beta. Hence, these terms can be used interchangeably. Overall, PKA C-beta is an essential enzyme that promotes various signaling pathways and is critical to maintaining cellular homeostasis.

SpecificityThe total PRKACB present in the organism can be detected using the PRKACB antibody. This antibody is capable of identifying endogenous levels of PRKACB, providing valuable insight into the activity of this protein. By detecting changes in PRKACB levels, researchers can identify potential molecular targets for therapeutic intervention. Highly similar content can be generated by rearranging the information of the original text, conveying the same meaning in a different way.

ApplicationThe dilution range for Western Blot (WB) is 1:500-1:2000, for Immunohistochemistry (IHC) is 1:50-1:200, for Immunofluorescence/Immunocytochemistry (IF/ICC) is 1:100-1:500, and for ELISA (peptide) is 1:20000-1:40000. These recommended dilution ranges ensure optimal experimental results and accuracy. It is important to dilute the samples properly in order to obtain reliable and consistent data.

Target

Background

Mediates cAMP-dependent signaling triggered by receptor binding to GPCRs. PKA activation regulates diverse cellular processes such as cell proliferation, the cell cycle, differentiation and regulation of microtubule dynamics, chromatin condensation and decondensation, nuclear envelope disassembly and reassembly, as well as regulation of intracellular transport mechanisms and ion flux. Regulates the abundance of compartmentalized pools of its regulatory subunits through phosphorylation of PJA2 which binds and ubiquitinates these subunits, leading to their subsequent proteolysis.

Tissue specificityIsoform 1 is most abundant in the brain, with low level expression in kidney. Isoform 2 is predominantly expressed in thymus, spleen and kidney. Isoform 3 and isoform 4 are only expressed in the brain.
Posttranslational modificationAsn-3 is partially deaminated to Asp giving rise to 2 major isoelectric variants, called CB and CA respectively.
