NF-kappa-B exists as a homo- or heterodimeric complex consisting of RELA/p65, RELB, NFKB1/p105, NFKB1/p50, REL, and NFKB2/p52. These dimers bind to specific DNA sequences called kappa-B sites, with each dimer having its own unique affinity and specificity for different kappa-B sites. Depending on the dimer combination, NF-kappa-B can function as a transcriptional activator or repressor.
The activity of NF-kappa-B is tightly regulated through post-translational modifications, subcellular localization, and interactions with cofactors or corepressors. In its inactive state, NF-kappa-B complexes are sequestered in the cytoplasm by members of the NF-kappa-B inhibitor (I-kappa-B) family.
