Description | The RNA polymerase II, responsible for synthesizing messenger RNA in eukaryotes, includes the fourth largest subunit encoded by this gene. In yeast, this particular subunit is associated with the polymerase during suboptimal growth conditions and may serve a role in protecting against stress. Interestingly, the transcript from this gene includes a ribosomal pseudogene sequence within its 3' untranslated region. This information was derived from RefSeq in July 2008. |
| Other names | The DNA directed RNA polymerase II 16 kDa polypeptide, also known as RPB4 or subunit D, plays a crucial role in transcriptional regulation. This protein is responsible for catalyzing the RNA synthesis from the DNA template during mRNA transcription. Its amino acid sequence is highly conserved across different species and it is required for the efficient processing of pre-mRNA. In humans, RPB4 is encoded by the polr2d gene. This subunit also interacts with other transcription factors and co-activators to control gene expression. Its importance in the regulation of gene expression has been demonstrated in numerous studies and mutations in the RPB4 gene have been linked with various disorders. Therefore, studying this protein and its interactions with other factors is crucial for a better understanding of gene regulation and its role in various biological processes. |

| Application | For Western blotting, the recommended dilutions are between 1:500 to 1:2000. When using immunohistochemistry, dilutions of 1:50 to 1:200 are suggested. For ELISA assays utilizing peptides, a dilution range of 1:20000 to 1:40000 should be used. It is important to follow these recommended dilutions in order to achieve accurate and reliable results in your experiments. |

Background | DNA-dependent RNA polymerase catalyzes the transcription of DNA into RNA using the four ribonucleoside triphosphates as substrates. Component of RNA polymerase II which synthesizes mRNA precursors and many functional non-coding RNAs. Pol II is the central component of the basal RNA polymerase II transcription machinery. It is composed of mobile elements that move relative to each other. RPB4 is part of a subcomplex with RPB7 that binds to a pocket formed by RPB1, RPB2 and RPB6 at the base of the clamp element. The RBP4-RPB7 subcomplex seems to lock the clamp via RPB7 in the closed conformation thus preventing double-stranded DNA to enter the active site cleft. The RPB4-RPB7 subcomplex binds single-stranded DNA and RNA (By similarity). |
