| Other names | PDHK2, also known as Pyruvate dehydrogenase kinase isoform 2 or PDK2, is a mitochondrial enzyme responsible for the regulation of pyruvate dehydrogenase activity. This enzyme acts by phosphorylating the E1 alpha subunit of the pyruvate dehydrogenase complex, leading to its inactivation and preventing the conversion of pyruvate to acetyl-CoA. PDHK2 has been implicated in various metabolic processes such as gluconeogenesis, lipogenesis, and glucose oxidation. The gene encoding PDHK2 is located on chromosome 7 and is expressed predominantly in tissues with high energy demands such as heart, skeletal muscle, and brain. Dysfunction of PDHK2 has been associated with metabolic disorders such as diabetes and cancer. Therefore, understanding the regulation of PDHK2 activity may provide new therapeutic approaches for the treatment of these diseases. |
| Source | Rabbit |
| Specificity | Endogenous levels of total PDK2 can be detected using the PDK2 antibody. The antibody is designed to specifically recognize and bind to PDK2, allowing researchers to accurately measure its levels. By detecting endogenous levels of PDK2, scientists can better understand its function and role in various biological processes. The highly specific nature of the PDK2 antibody ensures that only the target protein is detected, providing reliable and accurate results. |

| Application | For Western blotting, the recommended dilution range is between 1:500 to 1:1000. For immunofluorescence and immunocytochemistry, the suggested dilutions are between 1:100 to 1:500. For ELISA using a peptide substrate, the optimal dilution is between 1:20000 to 1:40000. These dilution ranges have been determined based on our extensive testing and experimentation. Please follow these guidelines when conducting experiments to ensure accurate and reliable results. |

Target | |
Background | Kinase that plays a key role in the regulation of glucose and fatty acid metabolism and homeostasis via phosphorylation of the pyruvate dehydrogenase subunits PDHA1 and PDHA2. This inhibits pyruvate dehydrogenase activity, and thereby regulates metabolite flux through the tricarboxylic acid cycle, down-regulates aerobic respiration and inhibits the formation of acetyl-coenzyme A from pyruvate. Inhibition of pyruvate dehydrogenase decreases glucose utilization and increases fat metabolism. Mediates cellular responses to insulin. Plays an important role in maintaining normal blood glucose levels and in metabolic adaptation to nutrient availability. Via its regulation of pyruvate dehydrogenase activity, plays an important role in maintaining normal blood pH and in preventing the accumulation of ketone bodies under starvation. Plays a role in the regulation of cell proliferation and in resistance to apoptosis under oxidative stress. Plays a role in p53/TP53-mediated apoptosis. |
| Tissue specificity | Expressed in many tissues, with the highest level in heart and skeletal muscle, intermediate levels in brain, kidney, pancreas and liver, and low levels in placenta and lung. |
