PAI-1, also known as plasminogen activator inhibitor 1, is a serine/cysteine proteinase inhibitor that belongs to the serpin peptidase inhibitor clade E. It is encoded by the SERPINE1 gene and is also referred to as serpin E1. PAI-1 is primarily expressed in endothelial cells and acts as an inhibitor of the plasminogen activators, effectively regulating fibrinolysis. It functions by binding to and inhibiting both tissue-type and urokinase-type plasminogen activators, thereby preventing the conversion of plasminogen to plasmin. PAI-1 plays a crucial role in maintaining the balance between coagulation and fibrinolysis within the body. Dysregulation of PAI-1 has been associated with various pathological conditions, including thrombosis, atherosclerosis, and cancer. Therefore, understanding the function and regulation of PAI-1 is of great importance in elucidating the underlying mechanisms of these diseases.

Application

The appropriate dilutions for ELISA, IHC-P, IHC-F, and IF are as follows: ELISA should be performed at a dilution of 1:500-1000, IHC-P and IHC-F at a dilution of 1:400-800, and IF at a dilution of 1:100-500. Please note that for paraffin sections in IHC, antigen retrieval is required.

PAI1 (plasminogen activator inhibitor 1) was initially discovered in human endothelial cell and rat hepatoma cell 3 cDNA libraries in the year 1986 by Pannekoek and Ginsburg, respectively. Falling into the serpin family of serine protease inhibitors, PAI1 possesses the capability to inhibit both tissue type plasminogen activator (tPA) and urokinase type plasminogen activator (uPA). An elevated level of PAI1 has been linked to an increased risk of thromboembolic disease. On the other hand, PAI1 deficiency may indicate an inherited autosomal recessive bleeding disorder.
