Description

eNOS, or endothelial nitric-oxide synthase, is an enzyme of great significance within the cardiovascular system. Its primary function is to facilitate the production of nitric oxide (NO), which plays a crucial role in regulating blood pressure, promoting vascular remodeling, and stimulating angiogenesis (1,2). To control its activity, eNOS undergoes phosphorylation at various sites. Among these sites, Ser1177 and Thr495 have been extensively studied (3). Multiple protein kinases, including Akt/PKB, PKA, and AMPK, contribute to the activation of eNOS by phosphorylating Ser1177 in response to various external signals (4,5). On the other hand, certain stimuli such as bradykinin and H2O2 promote eNOS activity by inducing phosphorylation of Ser1177 and dephosphorylation of Thr495 (6,7). Additionally, VEGF triggers the activation of eNOS through the dephosphorylation of Ser113. Notably, this dephosphorylation event can be obstructed by Cyclosporin A when stimulated by VEGF (8).

Other namesToday, I'd like to talk about cNOS. You may have heard it called by its full name, Constitutive NOS. Alternatively, you might have heard it referred to as EC-NOS, ecNOS, or Endothelial nitric oxide synthase. It's all the same thing, really! This enzyme is responsible for producing nitric oxide in our bodies. This is an important molecule that helps to regulate blood flow and blood pressure. Endothelial nitric oxide synthase is specifically found in endothelial cells, which line the inside of our blood vessels. That's why you might also hear it called Endothelial NOS or eNOS. No matter what name you use to refer to this enzyme, it's clear that it plays a vital role in maintaining cardiovascular health.
SourceRabbit
SpecificityThe NOS3 Antibody is capable of identifying total NOS3 present in the body at natural levels. The antibody has been developed to detect the endogenous levels of NOS3 and can be used for diagnostic and research purposes. By accurately identifying NOS3, this antibody can contribute to better understanding of the genetic and physiological factors that influence various diseases and conditions.

Target

Background

Produces nitric oxide (NO) which is implicated in vascular smooth muscle relaxation through a cGMP-mediated signal transduction pathway. NO mediates vascular endothelial growth factor (VEGF)-induced angiogenesis in coronary vessels and promotes blood clotting through the activation of platelets.

Tissue specificityPlatelets, placenta, liver and kidney.
Posttranslational modificationPhosphorylation by AMPK at Ser-1177 in the presence of Ca2+-calmodulin (CaM) activates activity. In absence of Ca2+-calmodulin, AMPK also phosphorylates Thr-495, resulting in inhibition of activity (By similarity). Phosphorylation of Ser-114 by CDK5 reduces activity.
Celluar localizationCytoskeleton;Cytosol;Golgi apparatus;Nucleus;Plasma Membrane;
